The role of central mechanisms in the anti-inflammatory effect of amitriptyline on carrageenan-induced paw edema in rats

Abstract: OBJECTIVE:The present study was designed to further investigate the effect of amitriptyline, a classical tricyclic antidepressant, on carrageenan-induced paw edema in rats.METHODS:First, amitriptyline was administered intraperitoneally (i.p.) at doses of 20, 40 and 80 mg kg-1, 30 min before subplantar injection of carrageenan. Second, amitriptyline was given intracerebroventriculary or intrathecally at doses of 25, 50 and 100 µg/rat, 30 min prior to carrageenan challenge. Third, the effect of adrenergic recept… Show more

“…34 Another study by Hajhashemi V et al, reported the antiinflammatory effect of amitriptyline in carrageenan induced paw edema in rats. 35 In present study authors found that acetic acid induced writhes were decreased in amitriptyline group, which signifies the peripheral analgesic action of amitriptyline. This finding also strengthens the above mentioned studies both direct and indirect way.…”

“…There is accumulating evidence from studies in rodents that TCAs, including the drug amitriptyline, have anti-inflammatory effects on various organ systems (Abdel-Salam et al 2003;Hajhashemi et al 2010) that often correlate with a decrease in the expression of cytokines, such as IL-6 or TNF-α (Tai et al 2006). While TNF-α is classically regarded as a pro-inflammatory cytokine, IL-6 can have either pro-or anti-inflammatory effects (Pedersen & Febbraio, 2008;Leonard & Maes, 2012).…”

“…The method of inflammation used in the present study was similar to that described in our previous work (Hajhashemi et al, 2010;Sadeghi et al, 2011). At first, the animals were pretreated with the extract (200, 500 and 750 mg kg À1 , p.o.)…”
