Screening of wild type Streptomyces isolates able to overproduce clavulanic acid

Abstract: The selection of new microorganisms able to produce antimicrobial compounds is hoped for to reduce their production costs and the side effects caused by synthetic drugs. Clavulanic acid is a β-lactam antibiotic produced by submerged culture, which is widely used in medicine as a powerful inhibitor of β-lactamases, enzymes produced by bacteria resistant to antibiotics such penicillin and cephalosporin. The purpose of this work was to select the best clavulanic acid producer among strains of Streptomyces belongi… Show more

“…Those were oleandomycin, 2-piperidinone and derivatives of either erythromycin or tylonolide. S. variabilis is reported for the production of L-Glutaminase, imunosuppresive, clavulanic acid, glycoside antibiotic and 1-hydroxy-1-norresistomycin (Abd-Alla et al, 2013, 2016; Marques et al, 2014; Ramalingam and Rajaram, 2016). …”

“…Because of the clinical importance of this compound, the increase in CA production has been the focus of several studies. Strategies to enhance CA production include optimization of batch or fed-batch operation [ 92 , 93 ], temperature [ 94 , 95 , 96 ], agitation and aeration [ 60 , 93 ], and medium composition [ 70 , 94 , 95 ], as well as the selection of new microbial strains [ 97 ].…”

“…Soybean derivatives (flour, protein isolate, and meal) have been used as nitrogen sources for CA production [ 7 , 67 , 70 , 94 , 95 , 97 , 106 , 107 ]. To perform a screening of medium constituents for fermentative CA production by S. clavuligerus , Rodrigues et al [ 70 ] performed fermentations using soybean protein isolate (SPI) and soybean flour (SF) as the primary nitrogen source, and obtained higher CA concentration (437 mg/L) with the former ingredient.…”

“…DAUFPE 3060, kindly provided by the Microorganism Collection of the Department of Antibiotics of the Federal University of Pernambuco, Recife-PE, Brazil, was recently submitted to molecular identification that revealed 96% identity of the 1,550 bp sequenced fragment with that of the strain Streptomyces variabilis 7525 (accession number: gi|341793371|JN180216.1); therefore, it was identified as Streptomyces variabilis DAUFPE 3060. 24 The microorganism was stored in cryotubes using 10% (v/v) glycerol as cryoprotectant at 2708C. Before its use in extractive fermentations, optical microscopy observations and cell viability tests were done to ensure that its characteristics had been preserved during cryopreservation.…”
