Optimization of media components for enhanced production of streptococcus phocae pi80 and its bacteriocin using response surface methodology

Abstract: The standard MRS components were optimized using response surface methodology for increasing yield of Streptococcus phocae PI80 viable cells and its bacteriocin. The highest amounts of bacteriocin activity and viable cells were recorded from prediction point of optimized MRS medium and achieved two fold higher (33049.8 AU.mL-1 and 14.05 LogCFU.mL-1) than un-optimized counterpart

“…The strain Streptococcus phocae PI80 was isolated previously from the gut of the Indian white shrimp [15,16,25]. TCBS was used for enumerating Vibrio parahemolyticus (MTCC-451).…”

“…The upper and lower limit of each parameter was specified according to OFAT optimization, as mentioned earlier. The value of α was selected based on a rotatable hypothesis for constant variance at points that were equidistant from the center point and supply equal precision of response evaluation in any direction of the design [ 34 ]. The CCD matrix was constructed with 30 runs.…”

“…To observe the emulsion stability, the emulsification index (E) was calculated as: [(volume of the emulsion layer × total volume −1 ) × 100]. As xanthan gum is one of the most widely used commercial bacterial EPS sources regularly used as an emulsifier [ 34 ], the emulsifying property of EPS was compared to other natural standard bacterial EPS xanthan gum (Sigma), along with synthetic surfactant Tween 20 (Sigma).…”

“…phocae PI80 is a potent probiotic strain, which was isolated from the gut of shrimp Penaeus indicus [14]. During the fermentation period, it produced some metabolites including exopolysaccharide and bacteriocin [17,18]. Moreover, S. phocae PI80 was found to exhibit potent probiotic role against vibriosis in shrimp Penaeus indicus and Aeromonas infection in fishes Cyprinus carpio and Tilapia mossambicus [19,20].…”
