The cytotoxic and growth inhibitory effects of palladium(II) complexes on MDA-MB-435 cells

Abstract: The antitumorigenic potential of two palladium(II) complexes, [Pd(ca2-o-phen)Cl2] – C1 and [Pd(dmba)(dppp)Cl] – C2, was evaluated, using MDA-MB-435 cells, a human breast adenocarcinoma cell-line that does not express the estrogen receptor α (ER−). Growth inhibition and induced alterations in cell-morphology were analyzed. The sulforhodamine B test showed that, compared to control cells, both C1 and C2 significantly inhibited (p < 0.5) cell growth. The maximum effect with both was achieved with 1 μM complexes, … Show more

“…Results showed that the manganese complex generated a significant decrease in the IC 50 for all the tested malignant cell lines when compared to curcumin alone, especially for the HOS and the A375 cell lines . Due to the structural similarity between Pt 2+ and Pd 2+ complexes, many authors have investigated the potential anticancer activity of palladium 2+ complexes and have demonstrated that they are cytotoxic to tumor cells . Generally, the complexes of Pd 2+ showed lower antitumor activity than cisplatin due to their more chemically labile nature .…”

“…Haematoxylin-eosin (HE) staining of cells for morphological studies was performed as previously reported[22], with modifications. After the incubation of cells plated on sterile microscope cover glass in Petri dishes, they were washed with 0.1 M PBS, pH 7.4, fixed with 70% acetone, dehydrated with ethanol series and HE staining was performed.…”

“…Moderate activity was observed for the remaining molecules. Complex 3h was identified as the highest tumour inhibiting molecule among all its derivatives and its cytotoxic potential was found to be 0.05 µM against 0.017 µM of a wellknown tumour inhibiting drug cisplatin [23]. The highest cytotoxic property of this molecule is due to its excellent symmetric arrangement [24].…”
