Isolation and characterization of genes functionally involved in ovarian development of the giant tiger shrimp Penaeus monodon by suppression subtractive hybridization (SSH)

Abstract: Suppression subtractive hybridization (SSH) libraries between cDNA in stages I (previtellogenic) and III (cortical rod) ovaries of the giant tiger shrimp (Penaeus monodon) were established. In all, 452 ESTs were unidirectionally sequenced. Sequence assembly generated 28 contigs and 201 singletons, 109 of which (48.0%) corresponding to known sequences previously deposited in GenBank. Several reproduction-related transcripts were identified. The full-length cDNA of anaphase promoting complex subunit 11 (PmAPC11;… Show more

“…Along with PGE 2, and PGF 2α , PGD 2 also has been identified in the rice field crab Oziothelphusa senex senex [13]. The presence of PGE 2 has been detected in the muscle and hemolymph of shrimp Penaeus monodon [14].…”

“…Prostaglandin D 2 (PGD 2 ), PGE 2 and PGF 2α have been detected in the fresh water field crab Oziotelphusa senex senex [20], while PGE 2 , thromboxane B2 (TXB 2 ) and 6-keto-PGF 1α have been reported in C. maenas [15]. In addition, PGE 2 has been identified in hemolymph, muscle and ovary of domesticated P. monodon [21], but the presence of PGF 2α has not been reported in this species.…”

“…Furthermore, injection of PGE 2 and PGF 2α into ovaries of O. senex senex significantly increased the number and the diameter of the oocytes in a dose-dependent manner [20]. In domesticated P. monodon , the amounts of PGE 2 in ovaries and haemolymph increased along with developing ovary stages [21]. However, the correlation between prostaglandins and crustacean ovary development may be species specific, as the amounts of PGE 2 and PGF 2α were highest in ovaries stage I and continued to decrease until stage IV in M. japonicus [10].…”

“…Preechapho et al, isolated 109 known sequences and identified several reproduction-related transcripts in ovaries of the giant tiger shrimps (P. monodon) using SSH (Preechaphol, Klinbunga, Khamnamtongand, & Menasveta, 2010). Adisak et al, obtained 185 transcripts and uncovered 8 novel YHV-responsive genes from haemocyte libraries of P. monodon induced by yellow head virus after 24h and 48/72h (Prapavorarat, Pongsomboon, & Tassanakajon, 2010).…”
