The aim of this study was to estimate the genetic diversity in 74 RR soybean cultivars from different Brazilian breeding programs. Analyzes were based on multivariate statistical techniques from phenotypic characteristics and microsatellite molecular markers (SSR). Ten agronomic traits were used in the analysis of the Euclidean distance, Tocher's clustering, UPGMA clustering and principal component analysis. Eighty-six of 100 SSR primer-pairs studied were selected based on their polymorphism information content, and analyzed using Jaccard Coefficient and UPGMA clustering method. The cultivars were clustered into seven groups according to the UPGMA and Tocher's methods, based on agronomic traits, while molecular analysis identified six groups. The phenotypic distances ranged from 0.46 to 9.79 and the dissimilarity measurements, based on SSR molecular markers, ranged from 0.07 to 0.73. Both results from agronomic traits and molecular markers showed that there is genetic variability among the RR cultivars and that the Monsanto breeding program has the most divergent germplasm. The analyzed agronomic traits and the chosen SSR markers were effective in assessing the genetic diversity among genotypes, besides proving to be useful for characterizing genetic variability of soybean germplasm.

ABSTRACT. The narrow genetic base of soybean makes cultivar characterization based on morphological descriptors difficult; this characterization is mainly done for registration and protection. Correct characterization of cultivars could be achieved through molecular markers, since the frequencies of each allele in the population are known. Consequently, we developed a molecular characterization method and initiated the construction of a molecular database for soybean cultivar identification. Thirty-two soybean cultivars were analyzed with 48 fluorescent-labeled microsatellite markers. The reactions were carried out in singleplex, and genotyping in quadriplex, using a capillary electrophoresis system in an automated sequencer. Probabilities of random identity and probabilities of random identity exclusion were calculated through estimated allele frequencies. A characterization profile was considered when the probability of random identity exclusion was equal or superior to 99.9999%. All steps of the experiment were doubled, using two independent sets of the same cultivar to evaluate the reproducibility of the method. A set of 13 microsatellite markers identified all 32 cultivars with 99.9999% certainty. The method was efficient and precise, with high reproducibility for cultivar characterization. These data are the beginning of a molecular database for soybean, and they can be used for cultivar characterization for registration and protection purposes and for cultivar identification in cases of intellectual property enforcement.

Resumo -O objetivo deste trabalho foi validar 19 marcadores microssatélites para resistência do trigo à giberela, em uma população não estruturada. Foram utilizados marcadores moleculares descritos na literatura como flanqueando QTLs de resistência à giberela em trigo, nos cromossomos 3B, 5A e 6B. Foram avaliadas 96 linhagens e cultivares de trigo quanto à severidade da infecção por giberela, em dois anos de avaliação. As linhagens e as cultivares foram genotipadas com 19 marcadores microssatélites. Os dados obtidos foram analisados pelo teste de Tukey e pelas análises de correlação, regressão linear simples e regressão múltipla; também foi estimada a eficiência de seleção dos marcadores moleculares. A severidade da doença variou de 1,95 a 41,3%, na média dos dois anos. Foram validados os QTLs nos três cromossomos avaliados. Os marcadores Xgwm389, Xgwm533, Xbarc180, Xbarc24, Wmc397, Xbarc101 e Wmc398 foram associados significativamente à resistência do trigo à giberela, tendo sido identificados alelos de resistência e de suscetibilidade. Validation of molecular markers for wheat resistance to Fusarium head blight in Brazilian wheat genotypesAbstract -The objective of this work was to validate 19 microsatellite markers for wheat resistance to Fusarium head blight (FHB), in an unstructured population. Molecular markers described in the literature as flanking QTLs of FHB resistance in wheat on chromosomes 3B, 5A, and 6B were used. Ninety six wheat lines and cultivars were evaluated for severity of FHB infection in two evaluation years. The lines and cultivars were genotyped with 19 microsatellite markers. The obtained data were analyzed by Tukey's test, and by correlation, simple linear regression, and multiple regression analyses; the efficiency of selection with molecular markers was also estimated. Disease severity ranged from 1.95 to 41.3% in the mean of the two years. QTLs in the three evaluated chromosomes were validated. The markers Xgwm389, Xgwm533, Xbarc180, Xbarc24, Wmc397, Xbarc101, and Wmc398 were significantly associated with FHB resistance in wheat, and alleles for susceptibility and resistance were identified.
