Novel Snake Venom Proteins Cytolytic to Cancer Cells in Vitro and in Vivo Systems

Abstract: Cancer cell inhibitors, named Atroporin and Kaotree, having molecular weights of 35 kDa and 6 kDa have been isolated from the venoms of Crotalus atrox and Naja naja kaouthia, respectively, by fractionation on high pressure liquid chromatography. The purified Atroporin and Kaotree showed killing effects on various types of human (breast, colon, liver, ovary, etc.) and animal cancer cells in concentrations as low as 0.5µg/ml, and having no effect on normal mouse kidney, liver, spleen, and erythrocytes up to 5.0µ… Show more

“…on adenocarcinoma cells. Thereafter many reports have established the anticancer potential of venoms of different species of Elapidae, Viperidae, Crotalidae snakes [102][103][104][105][106][107] and Hydrophis spimlis sea snake [108,109] and assigned it to phospholipase activity. Scorpions venom has been used by traditional and folk medicine in India, China, Africa and Cuba.…”

“…Studies have shown that venoms from snake species posses lytic activity against tumoral cells (Lipps 1999;Silva et al 2002) and antibacterial action (Stilles et al 1991) in vitro.…”

“…This was confirmed by MTT assay, the release of LDH is considered to be of necrotic pathway and the treated cells showing very less levels of LDH after 24 h suggests that the event of cell death is by apoptosis. Lipps observed similar results for the two protein fractions from snake venom, Atroporin and Kaotree on SP/2 cell line [24]. Moreover, Greel and Vaux [25] have suggested that a cancer cell treated with a potentially lethal toxin will not die because of the direct effects of toxin, but will activate its suicide mechanism to die earlier by apoptosis.…”
