Intra-peritoneal administration of Ecballium elaterium diminishes postoperative adhesions

Abstract: PURPOSE:To evaluate the effects of Ecballium elaterium (EE), Elaterium officinarum, in postoperative intraperitoneal adhesions in rats. METHODS:Thirty rats were divided into three groups and underwent midline laparotomy under 35 mg/kg ketamine and 5 mg/kg xylazine anaesthesia. In group 1 (n=10), the sham operation group, the abdominal walls were closed without any process. In group 2 (n=10), the control group, the antimesenteric border of the ceacum and the corresponding parietal peritoneum were abraded with d… Show more

“…Rats in the control group underwent sham laparotomy only, while sepsis and treatment groups were subjected to CLP after laparotomy; 2.5 mg/kg Ecballium elaterium extract and 2 cc saline were administered intraperitoneally (treatment group and sepsis group, respectively) as previously reported [10,11] and then incisions were closed by proper suturing of the peritoneum and skin layers. At post-operative 24 h, the animals were sacrificied and the brains were removed.…”

“…Adhesion may cause infertility, formed when oxygen radicals produced by cells break down lipid-containing structures, such as plasma and cell membranes, in the inflammatory response. [10,11,12] Tissue damage is used to assess the severity of inflammation and adhesion. Another parameter used to evaluate adhesion is hydroxyproline (OH-p), a hydroxylated derivative of the amino acid proline, which is abundant in collagen.…”

“…The fresh juice itself is used in treatment of sinusitis and jaundice [13,15]. Many studies have reported that EE also has antibacterial, antifungal, immunomodulatory, adhesion preventive effects, and also beneficial effects on sepsis-induced lung injury [8,9,15,16]. The understanding of underlying mech- …”

“…The insoluble materials were removed by vacuum filtration through Whatman paper. The ethanol in the extract was subsequently removed by evaporation and the extract was freeze dried at −80 • C. The extract was weighed and dissolved in sterile isotonic solution at a concentration of 100 mg/mL and stored in the refrigerator at 8 • C until its use for animal experiments [9].…”
