Light is one of the factors that influence the germination and initial establishment of orchids under in vitro cultivation. This study evaluated the effect of different light sources on these stages in in vitro cultivation of Schomburgkia crispa Lindl. After sowing in an aseptic environment, we stored the cultures in a screened greenhouse (natural light) or in a growth room with the following light sources: 3,000 K yellow LED; 6,500 K white LED [1]; 6,500 K white LED [2]; or 6,500 K white fluorescent lamp (control). We assessed germination percentage and initial seedling establishment at 45 and 90 days after sowing. Light did not influence the germination of S. crispa. However, the use of 3,000 K LED provided a faster initial establishment of S. crispa when compared to the other light sources, also presenting lower seedling mortality. Thus, the light source 3,000 K LED is a potential substitute for the 6,500 K fluorescent lamps and LEDs used in growth rooms in in vitro culture laboratories.

Passiflora edulis Sims, besides having economic and social importance, is included in the medicinal plant list that is considered of interest by the Unified Health System (Sistema Único de Saúde -SUS). Due to its high nutritional value, this species is widely used in food, cosmetics and pharmaceutical industries. It also contains carotenoids, as well as passiflorine and maracugine, which have sedative effects. The aim of this research was to assess P. edulis minimum growth conditions in different concentrations and combinations of MS culture medium and sucrose for in vitro conservation. The following variables were analyzed: Plant height; number of green and senescent leaves; and number of roots. Root height means were highest in MS media containing 30 g L -1 sucrose (8.33). With regard to plant height, the best results were obtained in MS/4 media with 15 g L -1 or 30 g L -1 sucrose, which enabled the maintenance of lower plants (5.35 and 4.91, respectively) with green leaves after 120 days in vitro culture. The MS/2 media supplemented with 15 g L -1 sucrose was the best alternative for in vitro maintenance of plants with appropriate height and number of green leaves (8.22 and 3.86, respectively). Thus, we may conclude that MS/2 with 15 g L -1 sucrose represents an efficient strategy for in vitro conservation of P. edulis plants for a culture period of 120 days and 100% survival after acclimatization.

Resumo Devido ao extrativismo, Orchidaceae apresenta elevada vulnerabilidade, necessitando de medidas para a conservação de suas espécies. Assim, objetivou-se determinar um protocolo para germinação e estabelecimento inicial de Cattleya nobilior, Cattleya lundii e Brassavola tuberculata quanto à desinfestação de sementes e a utilização de meios de cultura. Sementes foram desinfestadas em solução de NaClO a 0,8%, por 5 min ou 15 min, recebendo ou não tríplice lavagem. Em seguida, foram semeadas nos meios MS, MS 1/2, K ou VW. Posteriormente, foram transferidas para sala de crescimento com temperatura e fotoperíodo controlados. A porcentagem de germinação e o estabelecimento inicial das espécies estudadas foram avaliados 45 dias após a semeadura. Para todas as espécies, as sementes desinfestadas por 5 min e submetidas à tríplice lavagem apresentaram maiores %G. Dessa forma, recomenda-se para a germinação in vitro e desenvolvimento inicial de protocormos das espécies C. nobilior e B. tuberculata, a utilização do tempo de desinfestação de 5 min, seguido da tríplice lavagem e semeadura em meio MS 1/2 (%G = 100 e 72,8%, propágulos em estágios 3 e 2, respectivamente). Para C. lundii, a semeadura deve ser realizada em meio KC, com a desinfestação por 5 min e tríplice lavagem das sementes (%G = 95,8% e propágulos em estágio 2).
