ResumoO conhecimento do potencial germinativo otimiza a prática de germinação in vitro de Orchidaceae. Este trabalho avaliou o tipo e a concentração de soluções de tetrazólio sobre o tempo de reação e a identificação de sementes viáveis de orquidáceas. Foram utilizadas sementes das espécies Brassavola tuberculata Hook, Cattleya labiata Lindley var Amoema, Cattleya intermedia e Dendrobium antenatum e de dois híbridos primários de Cattleya labiata autumnalis x Cattleya labiata Lindley var. Alba. As sementes (0,01g) de cada espécie ou híbrido foram embebidas em 3 mL da solução aquosa ou da neutra de cloreto de 2,3,5 trifeniltetrazólio, nas concentrações de 0,0625%, 0,125%, 0,25%, 0,5% ou 1,0% e acondicionadas em ambiente desprovido de luz a 25±2 ºC, sendo avaliadas a cada 30 minutos para identificação do tempo necessário para a coloração dos embriões e posterior contagem do número de sementes viáveis por miligrama de sementes. Utilizou-se o delineamento experimental inteiramente casualizado, em esquema fatorial 6 x 2 x 5 (seis genótipos de orquídeas, dois tipos de soluções tetrazólicas e cinco concentrações) com 3 repetições. Os genótipos estudados responderam diferentemente no que se refere ao número de sementes viáveis por miligrama de sementes e quanto ao tempo necessário para a coloração dos embriões, sendo que C. intermedia não respondeu ao teste. Independentemente do tipo de solução, as concentrações só interferiram no tempo de reação para um dos híbridos. Recomenda-se, portanto, a utilização de solução neutra de tetrazólio a 0,5% para identificação de sementes viáveis dos híbridos de Cattleya labiata autumnalis x Cattleya labiata var. Alba e a utilização de solução aquosa a 0,125% para as espécies Brassavola tuberculata, Cattleya labiata var Amoema e Dendrobium antenatum. <p> O gladíolo é uma das plantas ornamentais mais cultivadas no Brasil, entretanto poucas são as informações científicas relacionadas à cultura. O objetivo desta pesquisa foi avaliar o efeito da adubação nitrogenada e tamanho do cormo no crescimento e floração de <em>Gladiolus grandiflorus </em>‘San Remo’. O experimento foi conduzido na área de Jardinocultura da Universidade Federal da Grande Dourados (UFGD) em Dourados – MS. O delineamento experimental utilizado foi inteiramente casualizado e os tratamentos dispostos em parcelas subdivididas, sendo as parcelas constituídas dos diâmetros dos bulbos (grandes entre 40 e 55 mm; médios entre 20 e 40 mm e pequenos inferior a 10 mm), e as subparcelas de três doses de nitrogênio (30, 50 e 70 kg ha-1), com 10 repetições. Foram avaliados o número de folhas, o comprimento da folha, o início da floração, o número de botões por espiga floral, a durabilidade da flor, a duração da floração e o número de cormilhos. As hastes florais produzidas foram classificadas como Classe III. A adubação nitrogenada foi mais eficiente para o crescimento de cormos de tamanho médio e pequeno, sendo que doses superiores a 50 kg ha -1 promoveram os melhores resultados. Em relação à floração, cormos de tamanho grande e a adubação nitrogenada de 30 kg ha -1 atrasaram seu início. Recomenda-se a utilização de cormos de tamanho médio e adubação nitrogenada de 50 kg ha -1 para o cultivo de <em>Gladiolus grandiflorus </em>‘San Remo’.</p>
The objectives of this study were to determine the optimal light conditions, agar concentration, and quantity of activated charcoal in asymbiotic media to improve the in vitro seed germination rate and early seedling development of the epiphytic orchids Dendrobium nobile Lindl. and Dendrobium phalaenopsis Fitzg. Two independent experiments with complete randomized designs were conducted. (1) Treatments were arranged in a split-plot scheme. Seeds in each sub-plot were exposed to one of four light conditions (dark, white fluorescent, red fluorescent + white fluorescent, or red fluorescent) and grown in one of four types of culture media (MS: Murashige and Skoog, ½ MS: half strength MS, K: Knudson C, and VW: Vacin and Went media). (2) Treatments were arranged in a 4 × 5 × 5 factorial scheme (four types of culture media: MS, ½ MS, K, and VW; five concentrations of agar: 0.0, 2.0, 4.0, 6.0, or 8 g L−1; and five concentrations of activated charcoal: 0.0, 1.5, 3.0, 4.5, or 6.0 g L−1). The highest germination rates and early seedling development were observed 45 days after seeding in the presence of white light for D. nobile and red + white light for D. phalaenopsis in MS and ½ MS culture media. Based on the findings of the present study, the use of MS and ½ MS culture media solidified with 4.0–8.0 g L−1 of agar and without activated charcoal is recommended for the optimal propagation of seeds and seedlings of these Dendrobium species.
The success in micropropagation of Dendrobium phalaenopsis Deang Suree is high, but when transplanted into the greenhouse, their survival is minimal. To increase survival in production in the present study it was evaluated the effect of intermediate acclimatization for 30 days in a grow room utilizing the following luminosity conditions: 1- white fluorescent light (B) (18.9µmol m-2 s-1); 2- white fluorescent light + red fluorescent light (GRO-LUX(r)) (BV) (14.85µmol m-2 s-1); 3- red fluorescent light (GRO-LUX(r)) (V) (9.45µmol m-2 s-1) and the control plants were accommodated directly in a greenhouse (162.0µmol m-2 s-1). After this the leaves were characterized anatomically and the plants transferred to the control greenhouse. It was evaluated survival percentage and final number of roots, and calculated the relations between the final and initial values of fresh weight, number of leaves, length and diameter of the largest pseudo bulb, number of pseudo bulbs and longest root length. Only plants submitted to red light, were statistically better than the control in relation to the survival percentage and in relation to fresh weight, while the control showed a higher number of roots that plants acclimatized in this luminosity conditions. Intermediate acclimatization, using red light or red + white light, is recommended for D. phalaenopsis Deang Suree.
RESUMO: Objetivou-se avaliar o crescimento da orquídea denphal, em função da luz, sistema de micropropagação e tamanho dos frascos de cultivo. O delineamento experimental utilizado foi inteiramente casualizado e os tratamentos arranjados em esquema fatorial 2 x 2 x 2 com cinco repetições. Plântulas cultivadas in vitro por 90 dias, a partir do início do período experimental, foram retiradas dos frascos, lavadas e avaliadas quanto ao número de folhas, altura da plântula, diâmetro do maior pseudobulbo, número de brotos e raízes, comprimento da maior raiz e da maior folha, e massa fresca. A seguir foram plantadas em substrato (esfagno rosa + fibra de coco) e transferidas para ambiente ex vitro. Com o intuito de investigar seus incrementos, após 90 dias, as plantas foram avaliadas quanto às mesmas características iniciais. Os diferentes espectros de luz não foram determinantes para o crescimento da planta. A utilização do sistema de ventilação natural em frascos de maior tamanho foi o fator que proporcionou condições favoráveis na promoção do crescimento, tanto no cultivo in vitro quanto no ex vitro do denphal.
Orchids are one of the groups of plants with the greatest commercialization potential. They have floral characteristics highly valued in the sector of flowers and ornamental plants. However, exotics species are the most commercialized. The native species are still little explored for their ornamental potential. Thus, this work describes the vegetative and floral morphological characteristics of Schomburgkia crispa Lindl. aiming at evaluating its ornamental potential. For such, seven individuals were chosen. The quantitative characters were evaluated in all plants, totaling fifty pseudobulbs and eighty-eight flowers. During flowering, the most representative plant was chosen for the visual evaluation of its morphological features through photographs. A floral diagnosis sheet based on photos of fresh flowers was prepared by keeping the maximum level of morphological details. At the end of the experimental period, averages and standard deviations were calculated for each character evaluated. The species S. crispa presented, on average, plants 46.17 cm long, 31.50 cm wide, and with 88 cm of diameter. For flowering characters, the species presented in general 16.50 flowers per plant, 14.67 per flower stem, and 4.66 flowers opened per day, with an average durability of 32 days. Thus, we conclude that the species S. crispa has a great ornamental potential for use in ornamental horticulture.
