Clinical hematology facilitates the diagnosis of disease and can act as a prognostic indicator of pathological conditions in fish. The aim of the present study was to evaluate hematological parameters of Nile tilapia (Oreochromis niloticus) subjected to different anesthetics and anticoagulants. Thirty apparently healthy fishes (average weight of 473 ± 35. 50 g and mean total length of 29. 33 ± 0. 37 cm), were selected from the local commercial fish farm in the Lages municipality (Santa Catarina, Brazil). The animals were randomly divided into three groups of 10. In two groups, anesthesia was induced with eugenol (70 mg·L -1 ) (EG) and Benzocaine hydrochloride (100 mg·L -1 ) (BG), respectively. Anesthesia was not administered to fish of the third group (CG/control group). Blood samples were obtained by venipuncture of the caudal vessels and placed into microtubes containing sodium heparin or Na 2 EDTA for further analysis. The results were analyzed by Sigma Stat for Windows, the paired t-test for significant differences between anticoagulants of the same group, and analysis of variance followed by the Tukey test for comparison of means between groups (p ≤ 0. 05). Most of the observed changes in the erythrogram were significantly higher for the anticoagulant heparin and benzocaine group in comparison to the control group. However, the values obtained for the leukogram were significantly higher for all groups subjected to the Na 2 EDTA anticoagulant, suggesting that heparin may cause cell clumping. The results suggest that the anesthetics under investigation effectively minimizes the effects of stress caused by handling and invasive procedures, and that the anticoagulant heparin causes less hemolysis in comparison to Na 2 EDTA for Nile tilapia. Thus, the hematological variations attributed to different anesthetic protocols and/or different anticoagulants should be considered for the species Oreochromis niloticus. O objetivo deste estudo foi avaliar os valores hematológicos de tilápias do Nilo (Oreochromis niloticus) submetidas a diferentes anestésicos e amostras obtidas com diferentes anticoagulantes. This study aimed to compare the body indexes and hematological characteristics between Astyanax bimaculatus males and females. Four hundred fish were randomly distributed into four polyethylene tanks (100 fish/unit) in a recirculation system and fed four times a day (3% of biomass). After 90 days, ten fish (five ♀ and five ♂) were removed to perform blood tests and to measure weight, height, total length, height/length ratio, condition factor and index determination: vicerosomatic (VSI), hepatosomatic (HSI), and gonadosomatic (GSI). The results showed a higher average weight (g) in females (12.32±0.71) compared to males (6.98±0.75), the same happened to height (cm) = (3.01±0.07) and (2.40±0.05), total length (cm) = (3.01±0.07) and (2.40±0.05), VSI (%) = (11.43±0.81) and (3.55±1.05), HSI (%) = (0.72±0.08) and (0.30±0.04), respectively. Mean corpuscular hemoglobin (pg) was higher in females (3.72±1.20) than in males (2.99±1.51). Regarding the number of thrombocytes (103.µL-1), there was an increase in males (25.71±3.91) compared to females (17.40±6.40).

Triploidy in the hematology of jundia juveniles (Siluriformes: Heptapteridae)

This study compared the hematological characteristics of diploid and triploid of jundia, Rhamdia quelen juveniles, an important freshwater fish cultured in south Brazil. Hematological morphometry of erythrocytes were determined in blood smears under a light microscope. The blood was used to measure the number of red blood cells (RBC) with a hemocytometer Neubauer chamber, and the numbers of white blood cells (WBC) and thrombocytes that were obtained using an indirect method. The results showed that triploidy increased (p < 0.01) the size and volume of the erythrocytes. Nevertheless, as expected, triploidy decreased (p < 0.01) the number of circulating erythrocytes, leucocytes and trombocytes in the blood of jundia. Moreover differential cell counts were different in diploids and triploids, suggesting that triploidy affects the number of cells and their proportion in blood. Lymphocytes were the most predominant cells in the differential counting of diploid fish (62.5%) while monocytes were predominant in triploid fish (49.6%). These results suggest performance differences between ploidies of jundia, and require future studies to evaluate the potential of triploid jundia in the culture conditions and resistance to infection.Keywords: Rhamdia quelen, triploid, diploid, blood cells. Triploidia na hematologia de juvenis de jundiá (Siluriformes: Heptapteridae)Resumo O presente estudo comparou características hematológicas de juvenis de jundiás Rhamdia quelen diploides e triploides, um importante peixe de água doce cultivado no sul do Brasil. A morfometria hematológica dos eritrócitos foi determinada em extensões sanguíneas sob microscopia óptica comum. O sangue dos animais foi também utilizado para mensurar o número total de eritrócitos em câmera de Neubauer, bem como os números totais de leucócitos e trombócitos pelo método indireto. Os resultados demonstraram que a triploidia aumentou (p < 0,01) o tamanho e volume dos eritrócitos. No entanto, como esperado, a triploidia reduziu (p < 0,01) o número de eritrócitos, leucócitos e trombócitos circulantes do jundiá. Além disto, a contagem diferencial de células sanguíneas foi diferente em diploides e triploides, sugerindo que a triploidia afeta o número das células, bem como sua proporção no sangue. Linfócitos foram as células predominantes na contagem de leucócitos de peixes diploides (62,5%), enquanto os monócitos foram os predominantes nos peixes triploides (49,6%). Estes resultados sugerem desempenhos diferentes entre as ploidias do jundiá e exige estudos futuros para avaliar o potencial dos jundiás triploides sob condições de cultivo e frente à infecção.Palavras-chave: Rhamdia quelen, triploide, diploide, células sanguíneas.
