Functional characterization and localization of AQP3 in the human colon

Abstract: Water channels or aquaporins (AQPs) have been identified in a large variety of tissues. Nevertheless, their role in the human gastrointestinal tract, where their action is essential for the reabsorption and secretion of water and electrolytes, is still unclear. The purpose of the present study was to investigate the structure and function of water channels expressed in the human colon. A cDNA fragment of about 420 bp with a 98% identity to human AQP3 was amplified from human stomach, small intestine and colon … Show more

“…Two AQP3 protein bands were detected. One of these appeared at ϳ27 kDa and represented the deglycosylated form of AQP3, whereas the other appeared at ϳ30 -40 kDa and represented a glycosylated form of AQP3 (32,33). This glycosylation is associated with the stability and intracellular translocation of AQP3 but has no influence on water permeability (1,8,35).…”

“…The second band was observed at approximately 50 kDa and represented the glycosylated form of AQP3. 13,14) Glycosylation has no influence on water permeability. [15][16][17] Therefore, in this study, both of these bands were accounted for when analyzing the total protein expression level of AQP3.…”

“…These are believed to represent non-glycosylated AQP3 and glycosylated AQP3, respectively. 15,16) The sum of these bands was analyzed as the protein expression level of AQP3. The protein expression levels of AQP3 in the colon increased over time following the administration of MgSO 4 , and at 8 h after administration, Rat faecal samples were collected at various times for up to 8 h beginning immediately after the administration of MgSO 4 , and faecal water content was measured.…”
