Concanavalin A-reactive nuclear matrix glycoprotein

Abstract: The binding capacity of concanavalin A (Con A) to condensed euchromatin and heterochromatin was investigated in chicken erythrocyte nuclei (CEN), mouse liver cells, Zea mays mays meristematic cells and Drosophila melanogaster polytene chromosomes after 4 N HCl hydrolysis to determine whether binding was preferentially occurring in bands and heterochromatin. “…The ability of ECF to bind to concanavalin A (Con‐A) extracted from Canavalia ensiformes was also investigated. Briefly, slides were incubated at room temperature with the Con‐A solution for 10 min, followed by a peroxidase solution for 10 min and by a peroxidase substrate solution containing 3,3′‐diaminobenzidine (Fluka Chemie, Switzerland), according to the protocols of Kiernan (35) and Vidal et al (36). This was performed because Con‐A–positive glycoproteins with physicochemical stability to acid hydrolysis that contributes to chromatin rheologic properties have been reported as being topologically located in heterochromatin and in condensed euchromatin even after a harsh HCl treatment, which removes histones (6, 36).…”
