Streptomyces sp. R18(6) and Streptomyces sp. 6(4) strains were evaluated for their ability to control brown spot and common root rot caused by Bipolaris sorokiniana in wheat crops. The antifungal activity of these isolates was tested using a double-layer assay and culture pairing at 28 °C. Physiological and enzymatic activity performed through siderophore, indole-3-acetic acid, nitrogen fixation and phosphate solubilization tests. The biocontrol of the disease and growth-promoting efficiency of wheat seedlings were assessed using in vivo assays in a greenhouse. In the culture pairing assays, both strains inhibited B. sorokiniana mycelial growth, while in the double-layer only Streptomyces sp. R18(6). Streptomyces sp. 6(4) produced auxin, siderophores, fixed nitrogen and solubilized phosphate, whereas R18(6) did not produce siderophores. In the greenhouse assays, strain R18(6) showed statistical differences in shoot dry mass and root dry mass compared with those of strain 6(4) in the presence of the phytopathogen (P ≤ 0.05), and these results were more evident when the environmental temperature was higher. In the absence of the phytopathogen, Streptomyces sp. 6(4) strain increased the root dry mass compared with that of the control during the same period. Therefore, these isolates can potentially control root rot and brown spotting and may promote the growth of wheat plants.

Actinobacteria secrete substances that limit or inhibit the growth of plant pathogenic fungi and may be used in the biocontrol of these microorganisms. The aim of this study was to characterize physiological and enzymatic activity of endophytic actinobacteria, evaluate their antifungal activity against Bipolaris sorokiniana root colonization, and evaluate their efficiency in promoting the growth of wheat seedlings. Antibiosis was analyzed using the double-layer method, the agar well diffusion test, and volatile metabolites. Physiological and enzymatic activity was evaluated through chitinase, glucanase, siderophores, indole-3-acetic acid, nitrogen fixation and phosphate solubilization tests. In vivo assays were evaluated by root colonization, biocontrol test and efficiency to promote the growth of wheat seedlings. From all isolates tested, 69.6% of them presented antifungal activity against at least one B. sorokiriana isolate. Among these, 17% of the isolates produced bioactive metabolites in the supernatant when grown in submerging culture. The highest production of bioactive metabolites was at 30°C, between 72 and 96 h of incubation. Three isolates produced volatile compounds, chitinase, glucanase, siderophores and exhibited nitrogen fixation, produced indole-3-acetic acid, efficiently colonized the root system of seedlings of two wheat cultivars. The best isolate [6(2)] showed, under the greenhouse, the capacity to promote an increased biomass and tillers per wheat plant.

Two in vitro assays were performed to verify the antagonistic activity of the isolates Bacillus licheniformis, B. subtilis, and B. subtilis + B. lichenformis against two isolates of Cff. All isolates of Bacillus spp. inhibited the growth of Cff isolates. Common bean seedlings showed no root colonization by the evaluated bacterial isolates.
