A simple, accurate, and sensitive method for the spectrophotometric determination of captopril in bulk and dosage forms is reported. The method is based on the bromination of captopril with excess solution of n-bromosuccinimide (NBS) in HCl acid medium. The excess NBS is pursued by the assessment of the residual NBS based on its ability to bleach the rhodamine B dye and measuring the absorbance at 555 nm. The amount of NBS reacted coincides to the drug content. The different experimental parameters influencing the development and stability of the color are precisely studied and optimized. Beer’s law is valid within a concentration range of 0.3–1.0 μg/mL with a correlation coefficient R2 = 0.991. The limit of detection 0.169 μg/mL is attained and relative standard deviation values for five replicated measurements of 0.3, 0.7, and 1.0 μg/mL captopril were between 0.53% and 2.03%. No interference is detected from prevalent additives found in pharmaceutical preparations. The proposed method is profitably put on to the determination of captopril in the tablet formulations with mean recoveries 98.91–101.27% and the results were statistically confronted with those of a reference method by applying Student’s t-and F-test.

Abstract.A new, simple and sensitive kinetic spectrophotometric method has been proposed for the determination of captopril (CPT) in pharmaceutical formulations. The method is based on oxidation of 3-methyl-2-benzothiazolinone hydrazone hydrochloride monohydrate (MBTH) by ferric chloride followed by its coupling with the drug to form green-yellow coloured product with absorbance maximum at 395nm. The concentration of CPT was calculated using the calibration equation for the rate data and fixed time methods. The linearity range was found to be 0.5-22.5 µg mL -1 for each method. The correlation coefficients were 0.9994 and 0.9971 for rate data and fixed time methods respectively. The proposed methods were applied successfully for the determination of CPT in pharmaceutical formulations. Statistical comparison of the results shows that there is no significant difference between the proposed and official methods.

Aim: This work deals with the simultaneous determination of captopril (CAP) and hydrochlorothiazide (HZ) in twocomponent solid dosage form, without prior separation, by three different techniques (simultaneous equation, absorbance ratio method, and first-order derivative method). Materials and Methods: This work was carried out on Shimadzu electron UV1800 double-beam UV-Visible spectrophotometer. The absorption spectra of reference and test solutions were carried out in 1 cm matched quartz cell over the range of 200-400 nm. Methanol and distilled water are used as solvent. Results: The first method is the application of simultaneous equation. Where the linearity ranges for both the drugs were 5-35 µg/ml. The second method is the determination of ratio of absorbance at 271 nm, the maximum absorption of HZ and isobestic wavelength 209 nm, the linearity ranges for both the drugs were 10-120 µg/ml The third method is the first-order derivative method, where the CAP shows wavelength at 222 nm and HZ shows at 340 nm, and the linearity ranges for CAP and HZ were 1-30 µg/ ml and 1-40 µg/ml, respectively. The proposed procedures were successfully applied for the simultaneous determination of both the drugs in commercial tablet preparation. The validity of the proposed methods was assessed by applying the standard addition technique where the percentage recovery of the added standard was found to be 99.52±0.214 and 99.00±0.165 using the simultaneous equation method, 99.76±0.684 and 99.58±0.279 using the graphical absorbance ratio method, and 99.45±0295 and 99.21±0.678 using first derivative method, for CAP and HZ, respectively. Conclusion: The proposed procedures are rapid, simple, require no preliminary separation steps, and can be used for routine analysis of both drugs in quality control laboratories.
