Development of a simple UV-spectrophotometric method for the determination of lansoprazole and study of its degradation profile

Abstract: A UV-spectrophotometric method is described for the determination of lansoprazole (LAN). The method is based on the measurement of the absorbance of LAN solution in acetonitrile at 281 nm. The system obeyed Beer's law over the concentration range of 1.25-25.0 µg/mL. The degradation behavior of LAN was investigated under dry heat treatment, UV-degradation, acid hydrolysis, alkali hydrolysis and oxidation; and found to degrade extensively under acid hydrolysis, alkali hydrolysis and oxidation. The method was app… Show more

“…The percent EE and DL were calculated as follows: EE (%) = (amount of drug entrapped in cochleates or liposomes/total amount of the drug initially added) Â 100; DL (μg/mL) = amount of drug entrapped in cochleates or liposomes/total amount of lipid. 7,13 Analysis of LSP was carried out using high-performance liquid chromatography (HPLC) as previously reported. 14 The isocratic mobile phase consisted of 10 mM phosphate buffer and methanol (45:55 vol/vol), then LSP detection was achieved at 285 nm.…”

“…6 As shown in Figure 1, LSP has an inherent sulfoxide group and reduction of this group takes place in exposure to acidic environment, resulting in degradation of LSP. 7 Without the protection via enteric coating, LSP is degraded very rapidly under acidic environments, while stable in neutral and alkaline conditions. 6 With this regard, strategies to protect LSP from the destructive effects of gastric acid are necessary.…”

“…The LPZ content was determined by weighing a specified amount of the LPZ-NCT cocrystals in a volumetric flask, dissolving it in methanol, and filling it to the desired volume with double-distilled water. The samples were then assayed using an ultraviolet-visible (UV-VIS) spectrophotometer at a wavelength of 283 nm [12].…”

“…The mass detector was operated in the electron-impact ionization mode (ionization energy, 70 eV). Molecular ion peaks were identified and possible structures were drawn on the basis of already reported data on decomposition pattern/chemical reactions of functional groups in OXCPM (14)(15)(16)(17)(18)(19).…”
