Caracterização da pureza de fosfatidilcolina da soja através de RMN de ¹H e de 31P

Abstract: Recebido em 31/7/07; aceito em 29/2/08; publicado na web em 22/9/08 CHARACTERIZATION OF SOYBEAN PHOSPHATIDYLCHOLINE PURITY BY 1 H AND 31 P NMR. A strategy is proposed to evaluate the purity of phosphatidylcholine from soybean lecithin, obtained by extraction or column chromatography, using the integrals ratio of 1 H NMR spectra. Integrals of methylene signals, around 1.3 and 1.6 ppm, are added and divided by the integral of the choline methyl groups, around 3.3 ppm. Before purification, a ratio of 19.68±1.37 w… Show more

“…For comparative purposes, purified lecithin was produced from crude soybean lecithin as previously described by Mertins et al 32 Briefly, 10 g of the granules of crude soybean lecithin was dispersed in ethyl acetate (100 mL) and ultrapure water (4 mL) was added under gentle shaking, leading to formation of a biphasic system. After decantation, the supernatant was discarded, and the lower phase was dispersed in acetone (60 mL).…”

“…The phosphatidylcholine content of the ASO mixture is described by approximately 75% distearoylphosphatidylcholine (DSPC:C 18:0), 12% dioleoylphosphatidylcholine (DOPC:C 18:1) and 8% dipalmitoylphosphatidylcholine (DPPC:C 16:0). The vesicle size reduction could be related to the mixture of lipids found in ASO, which can be responsible for several phase states, from cylinder to hexagonal ones (Mertins et al, 2008;Malheiros et al, 2010).…”

“…Liposomes, as model membranes, are also versatile tools to investigate a variety of physico-chemical properties or biological phenomena (De Lima, Morfim, Teixeira & Creczynski-Pasa, 2004). The amphiphilic nature of liposome components promotes the self-organization of bilayers, such as cell membranes (Mertins, Sebben, Schneider, Pohlmann & Silveira, 2008). Regarding the correlation of physico-chemical properties of liposomes and their biological activities, it would also be interesting to perform studies of unsaturated lipids, which can be used for preparing systems that resemble natural membranes, such as soybean asolectin (ASO), a mixture of phospholipids.…”
