Tissue culture parameters in sweet orange cultivars

Abstract: -The objective of this work was to establish tissue culture parameters for gene transfer in sweet orange cultivars. Epicotyl explants with different ages were cultured with 6-benzylaminopurine (BAP), kanamycin and hygromycin. Shoots were cultured with alpha-naphthaleneacetic acid (NAA) alone or in combination with indole-3-butyric acid (IBA). The requirement of BAP for shoot development was genotype-specifi c. Epicotyl explants from 35-day-old seedlings produced signifi cantly more shoots per explant in 'Pêra'… Show more

“…The present work corroborates these findings and further demonstrates that the optimal hormone concentrations are specific to cultivar, as it has been previously reported for juvenile explants (Bordón et al, 2000;Costa et al, 2004;Mendes et al, 2008).…”

“…), becoming the system inefficient and more complicated due to the need of micrografting. To our knowledge, only a few systematic studies evaluated the possible factors affecting in vitro rhizogenesis in citrus, which included auxins (Moreira-Dias et al 2000, Mendes et al 2008, temperature and photosynthetic radiation (Duran-Vila et al 1992). The role of polyamines (PAs) in this process has not been investigated, despite their relevance acquired during the last decade in relation to rhizogenesis (Couée et al 2004).…”

“…Similarly, highest shooting directly from explants ( Figure 1) (31.195%) was observed from in-vitro epicotyle ( Table 2). Mendes et al [9] also observed that epicotyl explants from 35-day-old seedlings produced significantly more shoots per explants in sweet orange variety 'Para'. No shooting from callus was observed on MS basal media+BAP 4 mg/l with/without IAA.…”
