Somatic embryogenesis and in vitro plant regeneration from pejibaye adult plant leaf primordia

Abstract: -The aim of this work was to evaluate a protocol for plant regeneration by means of somatic embryos obtained from isolated adult pejibaye leaf primordia, and to describe histological origin of embryos and morphogenetic response. Explants were cultivated in modified MS medium. Mesophyll parenchymatous cells originated meristemoids (preembryonic complex formation) induced with 7.1 µM BAP in the first two subculture periods. After polarized structures with 12.9 µM NAA and 3.55 µM BAP were formed in the third subc… Show more

“…Twenty seedlings were selected and cultivated in basal medium (MS -Murashige and Skoog 1962, modified by Almeida and Almeida 2006), followed by direct organogenesis induction on basal medium supplemented with α-naphthaleneacetic acid (NAA, 12.9 μM) and 6-benzylaminopurine (BAP, 3.55 μM). Subcultures were done twice, every 90 days, alternating with basal medium (Graner 2009).…”

“…Although palms are considered recalcitrant (Sarasan et al 2002;Steinmacher et al 2007), B. gasipaes in vitro regeneration through organogenesis and/or direct somatic embryogenesis has provided satisfactory results (Almeida and Kerbauy 1996;Almeida and Almeida 2006;Almeida et al 2012). However, further research is needed to optimize the propagation techniques.…”

“…Histochemical monitoring was used to determine the essential factors involved in the acquisition of embryogenic competence in cells and/or tissues (Kouakou et al 2007;Pinto et al 2011;Rocha et al 2011) and the distinction of embryogenic competence from organogenetic events. The establishment of in vitro plant regeneration, through direct organogenesis or somatic embryogenesis in the peach palm, has been described (Almeida and Kerbauy 1996;Almeida and Almeida 2006;Graner 2009). Both morphogenic events can occur simultaneously in this species (Graner 2009); however, a better understanding of the morphogenic processes is needed to reveal the precise factors that trigger different scenarios during in vitro plant regeneration.…”

“…Although a number of workers have investigated the in vitro multiplication of this species, including Almeida & Kerbauy (1996), Almeida et al (2005), Almeida & Almeida (2006), , micro-propagated seedlings are not easily encountered on a commercial scale due to high losses during their transfer to ex vitro conditions. Few workers have investigated acclimation of palm trees, and even fewer yet of peach palms.…”
