Genotypic Variability of Peanut Lines in Response to Water Stress, Based on Biochemical Descriptors

-Seven biochemical descriptors were used to estimate the genotypic variability of peanut in response to moderate water stress. Six genotypes, constituted by four lines and two cultivars, were grown in pots, each containing two plants. At 15 days after emergence (DAE), the treatment differentiation was carried out: Control-plants maintained with daily watering, and Stress-plants submitted to water stress by complete suspension of watering for 15 days. The experimental design was completely randomized with factorial scheme 6 x 2 (genotype x water treatments), with five replications. The biochemical variables evaluated were: catalase (CAT), ascorbate peroxidase (APX), guaiacol peroxidase (GPX), free proline, total carbohydrates, soluble proteins, and amino acids. Results obtained by biochemical analysis and estimation of genotypic variability indicated that proline is the most appropriate descriptor for selecting genotypes tolerant to water stress, which led to identification of L81V and L108V as promising lines for drought tolerance breeding program. VARIABILIDADE GENOTÍPICA DE AMENDOIM EM RESPOSTA A ESTRESSE HÍDRICO BASEADA EM DESCRITORES BIOQUÍMICOSRESUMO -Sete descritores bioquímicos foram utilizados para estimar a variabilidade genotípica de linhagens de amendoim submetidas a estresse hídrico moderado. Seis genótipos, sendo quatro linhagens e duas cultivares, foram cultivados em vasos, cada um contendo duas plantas e mantidas em casa de vegetação. Aos 15 dias após a emergência, procedeu-se a diferenciação dos tratamentos: Controle, as plantas mantidas com rega diariamente; Estresse, as plantas submetidas à suspensão total de rega durante 15 dias. O delineamento experimental foi inteiramente casualizado, com esquema fatorial 6 x 2 (genótipos x tratamentos hídricos), com cinco repetições. As variáveis bioquímicas avaliadas foram: catalase (CAT), ascorbato peroxidase (APX), guaiacol peroxidase (GPX), prolina livre, carboidratos totais, proteínas solúveis e aminoácidos. Baseado nos resultados das análises bioquímicas e de estimativa da variabilidade genotípica, a prolina demonstrou ser o descritor mais adequado para seleção de genótipos tolerantes ao estresse hídrico, contribuindo para indicar as linhagens L81V e L108V como mais promissoras, para um programa de melhoramento visando tolerância à seca. Drought is one of the environmental factors that most affects peanut cultivation in semi-arid regions, resulting in economic losses to growers. However, growth promoting bacteria are able to reduce water deficit damage in some plant species. In this context, this study aimed to evaluate the interaction of Bradyrhizobium strains reducing water stress effects on peanut genotypes by antioxidant enzymes activities, leaf gas exchanges and vegetative growth, as well as to determine the taxonomic positioning of strain ESA 123. The 16S rRNA gene of ESA 123 was amplified by PCR and sequenced by dideoxy Sanger sequencing method. An experiment was performed in greenhouse with three peanut genotypes (BRS Havana, CNPA 76 AM and 2012-4), two Bradyrhizobium strains (SEMIA 6144 and ESA 123), a mineral source of N and an absolute control (without N) under two water regimes (with and without irrigation). Seeds of peanut were sown and the plants were grown until 30 days after emergence. On the 20th day, the water deficit plants group had their irrigation suspended for 10 days. At in silico analyzes, ESA 123 presented 98.97% similarity with the type strain of B. kavangense. Leaf gas exchange was affected by water deficit; as well as alteration of antioxidant activities and reduction of vegetative growth variables. However, some plants inoculated with SEMIA 6144 and ESA 123 strains presented lower reductions and increment of some evaluated variables, mainly the ones inoculated with the ESA 123 strain, Bradyrhizobium sp. from the semi-arid region of Northeast Brazil. This data suggests beneficial effects of the peanut-Bradyrhizobium interaction in a water stress condition, specially with the ESA 123 strain.

As substâncias reguladoras podem auxiliar no crescimento inicial das plantas pela capacidade de interferir em mecanismos fisiológicos. Dessa forma, objetivou-se avaliar o crescimento inicial de plantas de amendoim após a aplicação direta nas sementes do bioativador Fertiactyl® LEG e do bioestimulante Stimulate®. O esquema experimental constou de 10 tratamentos arranjados em um esquema fatorial [(2 x 4) + 2], sendo o primeiro fator definido pelos produtos e o segundo fator as doses: 4,0 mL (T1), 8,0 mL (T2), 12,0 mL (T3),16,0 mL (T4) de Fertiactyl® LEG e 6,0 mL (T5), 12,0 mL (T6), 18,0 mL (T7), 24,0 mL (T8) de Stimulate® por kg de sementes, enquanto os tratamentos adicionais, a testemunha absoluta (T9) e relativa com 5 mL de água destilada kg-1 sementes (T10), no delineamento experimental inteiramente casualizado, com três repetições. Em seguida, as sementes tratadas foram distribuídas em sacos de polietileno (2 kg) de 15 cm de largura x 20 cm de comprimento, preenchidos com substrato composto de terra de superfície, terra de subsolo, compostagem e supersimples. Foram colocadas três sementes por saco em cada repetição. Aos 14 dias após a semeadura (DAS), realizou-se desbaste deixando-se uma planta por saco. Aos 30 DAS foram avaliados a altura de planta (cm), diâmetro da haste principal, número de folhas, massa seca de raiz, hastes e folhas e área foliar (dm2). Os dados avaliados foram submetidos a análise de variâncias do teste F e complementada por modelos de regressão para identificar o efeito de dose dos produtos sobre as características. Utilizou-se o programa estatístico SISVAR para análise estatística. O uso de bioativador Fertiactyl® LEG no tratamento de sementes de amendoim promove um aumento de 13,79% no diâmetro da haste principal e 30,68% na massa de matéria seca de raiz e resulta em maior crescimento e desenvolvimento da planta.
