The study was designed to assess the effects of in vitro selenium addition on intracellular hydrogen peroxide production by neutrophils from the milk and blood of dairy cows. Blood from 10 dairy cows and 20 milk samples from ϐive dairy cows were incubated with 0 mg (control) or 10μM of sodium selenite. Then, milk and blood neutrophils were submitted for evaluation of intracellular hydrogen peroxide production by ϐlow cytometry using 2',7'-dichloroϐluorescein diacetate as a probe. The selenium status of the animals was evaluated by determination of the blood glutathione peroxidase activity. The results of the present work showed that in vitro selenium supplementation leads to an enhancement in intracellular hydrogen peroxide production, which indicates an improvement in the bactericidal effects of blood and milk neutrophils even in cows with a selenium-adequate status. Thus, the present study showed that in vitro Se supplementation leads to an enhancement in intracellular hydrogen peroxide production, indicating an improvement in the bactericidal effects of blood and milk neutrophils in cows with Se-adequate status. A determinação da produção intracelular de peróxido de hidrogênio se deu por citometria de ϐluxo através da utilização do 2´,7´ diclorodihidroϐluoresceína diacetato como sonda. A mensuração do conteúdo de selênio foi avaliada pela atividade da glutationa peroxidase eritrocitária. Os leucócitos polimorfonucleares tanto sanguíneos quanto do leite apresentaram signiϐicativo aumento na produção intracelular de H 2 O 2 com a suplementação in vitro de selênio. Desta forma, o presente estudo apontou para aumento da produção intracelular de H 2 O 2 , indicando aumento da capacidade microbicida dos leucócitos polimorfonucleares sanguíneos e lácteos mesmo em animais com níveis adequados de selênio.

El objetivo de este estudio fue determinar el balance metabólico nutricional de Cu, Zn, Fe, Mn y Se, mediante la actividad sanguínea de SOD y GSH-Px y establecer la relación entre la concentración de Cu, Fe, Mn y Zn en el forraje y la actividad de SOD. Se tomaron 10 mL de sangre a 105 novillas seleccionadas en 15 rebaños lecheros de Caldas, Colombia (5º4' N y 75º3' O) y se tomaron muestras de forrajes para analizar Cu, Fe, Mn y Zn. El promedio de la actividad de SOD fue 1.390±1.299 U g-1 Hb, y estaba correlacionada con Cu, Mn y Fe en el forraje. La actividad promedio de GSH-Px fue 389±184 U g-1 Hb y fue observada una mayor frecuencia de valores deficitarios y bajo/marginales de 9%, habiendo sido más afectados los animales de zonas altas (>2.000 msnm). Bajo estas condiciones, estos resultados permiten señalar que la SOD es una enzima que puede emplearse como indicador del balance metabólico nutricional de Cu, Mn y Fe en bovinos a pastoreo, no está clara todavía su relación con Zn. La actividad de GSH-Px indica deficiencias en el balance metabólico nutricional de Se, en bovinos a pastoreo.

The objective of this study was to evaluate the effect of selenium (Se) supplementation on milk somatic cell count (SCC) in dairy cows. Twelve multiparous Holstein-Friesian cows were fed a diet containing a suboptimal Se concentration (<0.05 ppm, dry basis) starting 2 months before calving. Supplemented cows (n=6) received a single s.c. injection of barium selenate (1 ml/50 kg BW) 45 days prior to calving, whereas control group was kept unsupplemented. Twenty weeks after calving, two mammary quarters (right side) of each cow were challenged with 205,000 cfu/ml of Staphylococcus aureus (strain Newbould 305). Blood was collected bi-weekly until day 150 of lactation for the analysis of blood glutathione peroxidase (GPx1; EC 1.11.1.9) activity. To re-isolate the challenging pathogen and to evaluate SCC, aseptic milk samples were collected daily starting on the day of challenge, and finishing 7 days after inoculation. Unsupplemented cows had a lower activity of GPx1 through the experiment (P<0.001). Natural log SCC (lnSCC) was higher in unsupplemented than Se-supplemented cows (P=0.04), showing evidence of significance after 5 days. Selenium supplementation of dairy cows fed a diet containing a suboptimal Se concentration, resulted in higher blood activity of GPx1, and lower mean lnSCC after an intramammary challenge with Staph. aureus.
