Evaluation of an enzyme-linked immunoelectrotransfer blot test for the confirmatory serodiagnosis of human toxocariasis

Abstract: To improve the serodiagnosis of human toxocariasis, a sensitive and specific enzyme-linked immunoelectrotransfer blot (EITB-IgG) test was developed and evaluated using Toxocara canis larvae excretory-secretory antigens for detecting anti-Toxocara IgG antibodies. The EITB-IgG profile of toxocariasis was characterized by comparing 27 sera from patients with toxocariasis, 110 sera from healthy subjects and 186 sera from patients with other helminth diseases (ascariasis, ancylostomiasis, trichuriasis, enterobiasi… Show more

“…In the IgG-IB, sera from patients with toxocariasis recognized most of the protein bands, ranging from 29 to > 205 kDa. NUNES et al 17 found in the present study were not described by the other investigators 1,11,17,20 , and deserve to be further studied. Technical peculiarities used in TES production or the IB procedure may be the cause of such differences 1,17 .…”

“…For diagnosis, IgG is the best marker as it was detected in all patients with visceral, ocular and mixed forms of the disease. Low MW bands, described by other researchers 11,20 as specific, may be targets for IgG antibody detection in the diagnosis of toxocariasis. For monitoring patients after chemotherapy, IgG to > 205 kDa fractions, IgA to 29-38, 48-54, 81-93 kDa and IgE to 95-121 kDa may be useful.…”

“…Concentration of culture fluid containing ES antigen was achieved using PEG 20,000 in this study (Roldan and Espinoza 2009). Many authors reported the usage of ultra centrifugal devices with molecular weight cut off of 10,000 Da for concentration of culture fluid (Badley et al 1987;Bowman et al 1987 andKim et al 2008).…”

“…Thus, it is not possible to evaluate the performance of a test by comparing it to another with unquestionable results. For this reason, the ELISA-IgG test was chosen as a reference in this work based on the high degree of correlation demonstrated with the Western blot method (López et al 2005a, Roldán & Espinoza 2009, which is considered a confirmatory test (Magnaval et al 2001); additionally, the ELISA-IgG test is the internationally accepted diagnostic test. The k concordance index has been described as a way to comparatively analyse the agreement between two diagnostic tests with qualitative results (positive or negative).…”
