Mutations in the rpoB gene of rifampicin-resistant Mycobacterium tuberculosis strains isolated in Brazil and France

Abstract: We evaluated the mutations in a 193bp of the rpoB gene by automated sequencing of rifampicin (RMP)-resistant and susceptible Mycobacterium tuberculosis strains isolated from Brazil (25 strains) and France (37 strains). In RMP-resistant strains, mutations were identified in 100% (16/16) from France and 89% (16/18) from Brazil. No mutation was detected in the 28 RMP-susceptible strains. Among RMP-resistant or RMP-susceptible strains deletion was observed. A double point mutation which had not been reported befor… Show more

“…This contrasts with previous data showing that RIF-resistant isolates in Brazil exhibited rpoB mutation frequencies varying from 80–89% (Valim et al 2000, de Miranda et al 2001, Clemente et al 2008). Previous data on katG reported that mutations occur in approximately 60–85% of INH-resistant isolates (Rossetti et al 2002, Cardoso et al 2004, Hofling et al 2005, Clemente et al 2008) but, in our study, mutation of the katG gene was seen in 55% (65/118) of the INH-resistant isolates.…”

“…9,13,22,26,36,37 Mutations of codon 531, 526, and 516 have been reported as the most frequent mutations in the mutation hotspot region of the rpoB gene of MTB worldwide. 3,23,36 In the present study, the majority of mutations occurred at codon 526, and the mutation frequency (48.4%) observed was slightly higher than frequencies reported by China (40%), 36 Japan (35%), 16 and Korea (38%).…”

“…In addition, mutations in codon 533 have been reported for RMP-resistant and RMP-susceptible strains by Taniguchi et al [74]. However, in studies conducted with isolates from same regions in Brazil and France, this mutation was strongly involved with RMP resistance [75].…”
