Bioactivity of neolignans from fructus Schizandrae

Abstract: Fructus Schizandrae sinensis Baill, a traditional Chinese medicine, used as tonic and sedative, has been shown at the beginning of 70's to lower the elevated serum glutamic-pyruvic transaminase (SGPT) levels of patients suffering from chronic viral hepatitis. During past 20 years, a series of neolignans have been isolated and identified as effective principles. Pharmacological studies revealed that they increased liver protein and glycogen synthesis, antagonized liver injuries from CCl4 and thioacetamide. The … Show more

“…Treatment with Schisandrin C synthetic analogue ( DDB ) , significantly recovered both liver weight , TL and TP to a non -significant difference from control , although TP didn't return to the control value , but only improved by drug treatment . This probably necessitated a longer treatment period for complete recovery , because DDB treatment was proved in other situations to initiate both protein and glycogen synthesis in CCl4 -intoxicated rats and protected liver tissue by membrane stabilization (Li ,1991 ) .In the present results , CCl4 significantly increased both tissue TC and TG , while decreased PL contents .Treatment with DDB , significantly reversed this effect and recovered the three parameters near to normal control values. CCl4 was known to interfere with the hepatic PL synthesis (Recknagel et al , 1976 ) and to decrease liver plasma membrane PL content in both acute and chronic treatments (Camacho and Rubalcava ,1984 ) .…”

“…20,21) Pharmacological studies revealed that lignans isolated from Schisandra chinesis show anti-cancer, anti-hepatotoxic, anti-oxidative, and anti-inflammatory activities. [22][23][24] Gomisin A, for example, has several pharmacological activities, including anticancer and cancer chemopreventive potential in animal models. [25][26][27] Tumor necrosis factor-α (TNF-α) causes a hemorrhagic necrotic effect against implanted solid tumors.…”

“…We previously reported schisandrin B (SchB) as the first example of a specific inhibitor of ATR in DNA damage checkpoint signaling (10). SchB is the major lignan isolated from Schisandra chinensis, one of the most frequently used herbs in traditional Chinese medicines (11,12), and it has subsequently been revealed to be a mixture of gomisin N (GN) and -schisandrin (-Sch), which are the diastereomers of each other (Fig. 1).…”
