Introduction: Acute gastroenteritis (AGE) is one of the most common causes of morbidity and mortality, especially among children from developing countries. Human adenovirus (HAdV) and sapovirus (SaV) are among the agents that cause AGE. The present study aimed to detect and genotype HAdV and SaV in 172 fecal samples from children with AGE, collected during a surveillance study carried out in a low-income community in Belém, Pará, between 1990 and 1992. Methods: HAdV was detected by nested PCR, using primers Hex1deg/Hex2deg and NeHex3deg/NeHex4deg. SaV was assayed by reverse transcription PCR (RT-PCR), nested PCR, and quantitative PCR. The nucleotide sequence was determined by direct cycle sequencing. Results: Overall, 43% (74/172) of samples were positive for HAdV, of which 70.3% (52/74) were sequenced and classified as belonging to five different species, mostly A and F. For SaV, positivity was 5.2% (9/172) and genotypes GI.1, GI.7, GII.1, and GV.2 were detected. Conclusions: The present results reinforce the need for further studies to obtain epidemiological data about the circulation of these viruses in Brazil, especially in the Amazon Region, where data from the early 1990's are scarce. Furthermore, the study describes for the first time the detection of SaV genotypes GI.7 and GV.2 in Brazil, showing that these types circulated in the region more than 25 years ago.

Adenoviruses are the most common causative agents of epidemics of community acquired and nosocomial conjunctivitis. Adenoviruses are resistant to physical and chemical factors and they can spread easily. Due to these factors, rapid and accurate diagnosis is very important in adenoviral eye infections. In this study, it was aimed to investigate adenoviruses from conjunctival swab samples of 50 patients suspected of adenoviral conjunctivitis by two different cell culture and PCR methods. Conjunctival swab samples from patients suspected of adenoviral conjunctivitis were taken between March 2012-December -11, 15, 16, 17, 19, 20, 22 cause conjunctivitis, serotypes 2, 3, 4, 5, 7, 8, 10, 11, 19, 21, 22, 29, 34, 37, 53, 54, 56 cause epidemic keratoconjunctivitis and serotypes 3, 4 and 7 cause pharyngoconjunctival fever (5,7,8). Outbreaks of adenoviral infections are common (9). Due to remaining infectious at room temperature for weeks and high level of resistance to environmental factors, transmission of adenovirus may occur easily (9,10). As can be transmitted from person to person, transmission may occur through water, objects and medical devices. Therefore nosocomial adenovirus infections can be seen (10).Adenoviral infections are mainly diagnosed by direct methods such as cell culture, nucleic acid detection and antigen detection (10,11). Indirect diagnosis based on the serological methods are mainly preferred in epidemiological researches instead of diagnosing adenoviral infections because of their low sensitivity rates (11). Rapid diagnose in adenoviral eye infections, especially in outbreaks of nosocomial conjunctivitis is very important and essential for confirmation of clinical diagnosis, limiting the spread of infection, implementation of appropriate protective measures and prevention of inappropriate treatment (12). Promptly isolation of infected individuals is critically important for control of outbreaks in both community acquired and nosocomial outbreaks (13). In this study, it was aimed to investigate adenovirus from conjunctival swab samples of patients suspected of adenoviral conjunctivitis by cell culture and PCR methods. MATERIALS AND METHODSThis study was performed in Virology Laboratory, with approval of Local Ethics Committee approval (06/05/2010) and supported by Scientific Research Projects Department (TSU-11-3645 code number). Total of 50 patients admitted to Ophthalmology Clinic whom suspected of adenoviral conjunctivitis between March 2012-December 2012 were included in this study. Introduction: Highly contagious adenoviral conjunctivitis represents 15-70% of all conjunctivitis worldwide. Human adenovirus (hAdV) serotypes 3,4,7,8,19 and 37 contributes to 89% of all adenoviral conjunctivitis. Accurate and rapid diagnosis of adenoviral infections at serotype level could prevent misdiagnosis, spread of disease, unnecessary antibiotic use and increased treatment costs. Methodology: Sixty-two suspected viral conjunctivitis cases were recruited from November2013-January2015. Swabs collected from inferior palpebral conjunctiva and processed for viral culture (Hep2 cell line), immunofluorescence assay (IFA) and polymerase chain reaction (PCR) (targeting hexon gene). Serotype 3,4,7,8,19 and 37 identification was carried out with an optimized multiplex-PCR (based on hypervariable region of hexon gene) and confirmed by sequence analysis. Bayesian Latent Class Model (LCM) analysis was used to compare sensitivity and specificity of three tests. Results: Adenovirus was detected in 54.8% (34/62) of cases by combination of all three methods. Culture was positive in 23/34 cases (67.6%). PCR and IFA detected adenovirus in 24 (70.5%) and 21 (61.7%) cases respectively. LCM analysis revealed, sensitivity and specificity of PCR, Culture and IFA was 77.8% and 92.4%; 72.2% and 90.8%; 67.6% and 92.9% respectively. Serotyping by multiplex-PCR showed, two cases each were hAdV3 and hAdV4, 18 hAdV8 and two remained unidentified. Results of Multiplex-PCR and sequence analysis showed 100% concordance Conclusion: LCM analysis revealed, PCR is the most appropriate method for identification. Multiplex-PCR is a simple and rapid method (serotypes identification within two days); owing its short turnaround time and accuracy, it can be used as a diagnostic tool for surveillance of adenoviral keratoconjunctivitis.
