The myenteric plexus of the digestive tract of the wild mouse Calomys callosus was examined using a histochemical method that selectively stains nerve cells, and the acetylcholinesterase (AChE) histochemical technique in whole-mount preparations. Neuronal density was 1,500 ± 116 neurons/cm 2 (mean ± SEM) in the esophagus, 8,900 ± 1,518 in the stomach, 9,000 ± 711 in the jejunum and 13,100 ± 2,089 in the colon. The difference in neuronal density between the esophagus and other regions was statistically significant. The neuron profile area ranged from 45 to 1,100 µm 2 . The difference in nerve cell size between the jejunum and other regions was statistically significant. AChE-positive nerve fibers were distributed within the myenteric plexus which is formed by a primary meshwork of large nerve bundles and a secondary meshwork of finer nerve bundles. Most of the nerve cells displayed AChE activity in the cytoplasm of different reaction intensities. These results are important in order to understand the changes occurring in the myenteric plexus in experimental Chagas' disease.

Does Cyclophosphamide Play a Protective Role Against Neuronal Loss in Chronic T. cruzi Infection?

In this study, we verified the possible role of cyclophosphamide (CY) in protecting or not against neuronal losses in young and aged male Calomys callosus chronically infected with the MORC-1 strain of Trypanosoma cruzi through numerical quantification of neurons from the myenteric plexus of the colon and quantification of nitric-oxide concentration (NO) during the acute and chronic phase of infection. For this purpose, groups of young C. callosus were infected with the MORC-1 strain of T. cruzi. A group of infected animals received i.p. 0.2 mg/ml genuxal dissolved in distilled water treatment with CY. NO concentration in aged animals displayed reduced levels when compared to those found in young animals. No significant alterations in the number of neurons were observed in young animals, but for aged ones, a protective role of CY in reducing neuron loss was noted, in addition to enhancing the neuronal volume, area, and perimeter. These results suggest that CY administration, depending on the dose and time span, can act as a protective agent against neuronal losses.

Foi descrita a infecção experimental em Calomys callosus com uma cepa de Leishmania donovani chagasi de caso humano. Um grupo de 22 roedores foi inoculado por via intraperitoneal com 0,1 ml de um macerado de baço em salina, rico em amastigotas. Esses animais foram sacrificados três meses após as inoculações, tendo sido realizado: cultura "in vitro" em meio acelular (LIT e NNN) e esfregaços, corados pelo Giemsa, de fígado, baço, medula óssea e sangue; cortes histológicos corados com hematoxilina-eosina de fígado e baço. Os resultados para fígado e baço foram: 67% de positividade nas culturas "in vitro"; esfregaços ricos em amastigotas intra e extra celular (inclui medula óssea); reações teciduais traduzidas por hepatomegalia com proliferação das células de Kupffer; reação granulomatosa das áreas portais, esplenomegalia com reações granulomatosas, abundância de formas amastigotas. Os resultados para o sangue foram negativos em todas as investigações.
