-765 G>C POLYMORPHISM OF THE COX-2 GENE AND GASTRIC CANCER RISK IN BRAZILIAN POPULATION

Abstract: -Context -Genomic alterations play important roles in gastric cancer carcinogenesis. Cyclooxygenases (COX) are important enzymes in the maintenance of mucosal integrity and in pathological processes, mainly in inflammation and cancer. The -765G>C COX-2 polymorphism has been implicated in gastric cancer risk. Objective -To evaluate the COX-2 gene polymorphism as a predictor of gastric cancer risk. Methods -One hundred gastric cancer patients and 150 controls were enrolled from a Brazilian centre. Personal data … Show more

“…A total of 15 original papers that met the inclusion criteria were selected for the meta-analysis; they included an assessment of the association between COX - 2 rs20417 polymorphism and GC in 2848 cancer cases and 4962 healthy controls. [22–27,3139] These papers reported studies of Asian (11 papers), [22–26,31,32,3437] Caucasian (3 papers), [27,33,38] and American (1 paper) [39] (details shown in Table 1). The association between COX - 2 rs20417 polymorphism and GC susceptibility in H pylori positive and H pylori negative patients under the dominant model were available in 5 studies.…”

“…IL-8 c.-251A > T, IL-10 c-592C > A, and IL-10 c.-1082 A > G are the most studied germline risk variants, with GA susceptibility studies reported in countries such as Brazil, 39 , 40 , 41 , 42 , 43 Chile, 44 , 45 Colombia, 46 Mexico, 47 and Peru 48 ( Fig. 2 and Table 2 ).…”

“…Prostaglandin-endoperoxide synthase 2 (PTGS2) or Cyclooxygenase-2 (COX2) is an enzyme involved in inflammation and tissue damage responses, which could lead to cancer. Two SNPs rs20417 (c.-899G>C, C allele) and rs689466 (c.-1329A>G, A allele) increasing COX2 protein expression have been reported as potential susceptibility biomarkers [9][10][11][12][13][14]. On the other hand, epidermal growth factor receptor (EGFR), a tyrosine kinase receptor activated by the epidermal growth factor (EGF), is essential in laryngeal squamous epithelium differentiation, survival, and proliferation.…”
