Sporadic colorectal cancer (CRC) is a consequence of the accumulation of genetic and epigenetic alterations that result in the transformation of normal colonic epithelial cells to adenocarcinomas. Studies have indicated that a common event in the tumorigenesis of CRC is the association of global hypomethylation with discrete hypermethylation at the promoter regions of specific genes that are involved in cell cycle regulation, DNA repair, apoptosis, angiogenesis, adhesion and invasion. The present study aimed to investigate the epigenetic changes (DNA methylation) in 24 candidate genes in CRC. A total of 10 candidate hypermethylated (HM) and unmethylated (UM) genes were identified that may be useful epigenetic markers for non-invasive CRC screening. The five genes that had the highest average UM percentages in the control group were MLH1 (71.7%), DKK2 (69.6%), CDKN2A (68.4%), APC (67.5%) and hsa-mir-342 (67.4%). RUNX3 (58.9%), PCDH10 (55.5%), SFRP5 (52.1%), IGF2 (50.4%) and Hnf1b (50.0%) were the five genes with the highest average HM percentages in the test group. In summary, the present preliminary study identified the methylation profiles of normal and cancerous colonic epithelial tissues, and provided the groundwork for future large-scale methylation studies.
Parkinson's disease (PD) is characterized by a reduction in the number of dopaminergic neurons of the substantia nigra (SNpc), accompanied by motor and non-motor deficiencies such as respiratory failure. Here, our aim was to investigate possible neuronal communications between the SNpc and chemoreceptor neurons within the retrotrapezoid nucleus (RTN), in order to explain neurodegeneration and the loss of breathing function in the 6-OHDA PD animal model. Male Wistar rats received tracer injections in the SNpc, RTN and periaqueductal gray (PAG) regions to investigate the projections between those regions. The results showed that neurons of the SNpc project to the RTN by an indirect pathway that goes through the PAG region. In different groups of rats, reductions in the density of neuronal markers (NeuN) and the number of catecholaminergic varicosities in PAG, as well as reductions in the number of CO-activated PAG neurons with RTN projections, were observed in a 6-OHDA model of PD. Physiological experiments showed that inhibition of the PAG by bilateral injection of muscimol did not produce resting breathing disturbances but instead reduced genioglossus (GG) and abdominal (Abd) muscle activity amplitude induced by hypercapnia in control rats that were urethane-anesthetized, vagotomized, and artificially ventilated. However, in a model of PD, we found reductions in resting diaphragm muscle activity (Dia) and GG frequencies, as well as in hypercapnia-induced Dia, GG and Abd frequencies and GG and Abd amplitudes. Therefore, we can conclude that there is an indirect pathway between neurons of the SNpc and RTN that goes through the PAG and that there is a defect of this pathway in an animal model of PD.

Na maioria, essas alterações genéticas ocorrem em uma única célula somática, que então se divide e continua se desenvolvendo até formar um câncer. Mais raramente, quando a neoplasia maligna ocorre como parte de uma síndrome de câncer hereditário, as alterações iniciais são herdadas por meio de linhagem germinativa e, portanto estão presentes em todas as células do organismo (15) .Mutações e polimorfismos são duas alterações genéticas freqüentes. As mutações são representadas pela substituição de bases, alterações na organização ou no tamanho das seqüências, incorporação do DNA extracromossômico e alterações anafásicas ou da citocinese. Essas alterações estão associadas à freqüência de alelos heterozigotos presentes em menos de 2% da população (1) . Os polimorfismos genéticos são variações na seqüência de DNA que podem criar ou destruir sítios de reconhecimento de enzimas de restrição e parecem estar associados a apenas uma base. A freqüência de alelos heterozigotos para o polimorfismo genético ocorre em mais de 2% da população. Algumas dessas alterações ocorrerão em seqüências não codificadoras do gene, que na maioria dos casos não terão efeito em suas funções; outras ocorrerão em seqüências codificadoras, levando à produção de proteínas defeituosas. Deste modo, em alguns casos o polimorfismo genético pode aumentar a suscetibilidade ao câncer (11) .Mundialmente, o câncer colorretal foi a quarta neoplasia mais incidente tanto no sexo masculino, quanto no feminino. A sobrevida global em 5 anos é de 60% e não são observadas diferenças significativas entre países desenvolvidos e em desenvolvimento. No Brasil, o câncer de cólon e reto é a quinta causa de morte no sexo masculino e a terceira no sexo feminino (5) , tendo-se observado aumento consistente de suas taxas de mortalidade ao longo das últimas décadas. RESUMO -Racional -Cerca de 35% dos casos de câncer estão envolvidos com fatores advindos da dieta e de outros como o álcool, o fumo, a luz solar, agentes químicos e infecções por vírus. O câncer de estômago é o segundo tipo de câncer mais diagnosticado no mundo, sendo responsável por cerca de 9,9% de todos os diagnósticos e cerca de 12,1% das mortes. INTRODUÇÃOO câncer, segundo dados da Organização Mundial da Saúde, é diagnosticado em cerca de 11 milhões de pessoas por ano no mundo. Estudos sugerem que essa doença poderá atingir cerca de 16 milhões por ano em 2020 (29) . Anualmente, cerca de 7 milhões de mortes são provenientes de câncer, o que representa 12,5% do total de mortes no mundo (29) . Os cânceres de pulmão, colorretal e de estômago são os tumores mais comuns no mundo para homens e mulheres (29) .Indivíduos que apresentam câncer do sistema digestório relatam freqüentemente perda de peso em curto período de tempo (1,2) , dificuldade de alimentação, dor local, náuseas, vômitos, flatulência, sensação de plenitude precoce que contribuem para o agravamento da doença, dificultam os tratamentos propostos e, conseqüentemente, favorecem pior prognóstico (1,2,16) .O câncer de estômago é o segundo mais diagnosticado no mundo, sendo responsável por cerca de 9,9% de todos os diagnósticos e cerca de 12,1% das mortes. Sua incidência é maior em algumas regiões da China, Ásia e América do Sul (29) .Cerca de 35% destes cânceres estão relacionados a fatores advindos da dieta (16,19,25,30) , assim como do consumo de álcool, do uso de tabaco, da exposição à luz solar, de agentes químicos e infecções por vírus (9) .
