Feasibility of the new method for orchid in vitro rooting using liquid and chemical sterilized culture medium under different sucrose concentration

Abstract: The in vitro propagation of orchids is the only commercial large scale technique to obtain healthy and high quality plantlets with clonal origin. The use of new technologies in plant tissue culture systems could lead to efficiency increases and costs reduction of micropropagation systems. The main actual micropropagation system is based on semi-solid culture media solidified using agar, followed by sterilization using autoclaving, and cultivation under photomixotrophic conditions using sucrose as main source o… Show more

“…This is possible because no variation was applied in temperature or in the supplied sucrose, which participates in seedlings' respiration (Yang et al 2015). The global photosynthetic rate was signi cantly higher in GIB with immersion every 12 hours (0.74 µmol CO 2 g − 1 s − 1 ) than in the seedlings grown in semisolid media (0.27 µmol CO 2 g − 1 s − 1 ) because the gas exchange is reduced in semi-solid medium (Oliveira et al 2019). On average, global photosynthesis was twice as high in the bioreactors with liquid medium (0.54 µmol CO 2 g − 1 s − 1 ) compared to the semi-solid one (Table 4).…”
