Background: Lactic acid bacteria (LAB), many of which are probiotics, can produce health-promoting metabolites (postbiotics). Purpose: To assess the mechanism of antiproliferative action of postbiotics, post-fermentation media (PFM) and cell extracts (CEs) of several strains of LAB were studied against colon (Caco-2), and cervix (HeLa) cancer cell lines, as well as normal intestine (IEC-6) cells, were used as a comparison. Methods: Postbiotics of various LAB (n = 39) were screened for their antiproliferative activity. The effect of PFM and CEs on reactive oxygen species (ROS), mitochondrial membrane potential (MMP), ATP production, phosphatidylserine (PS) externalisation, and apoptosis-related caspases 3/7 and 9 activation was assayed. Results: PFM and CEs showed strong dose-dependent antiproliferative activity against Caco-2 cells, up to 77.8 ± 0.8% and 58.4 ± 1.6% for PFM and CEs, respectively. Stronger inhibitory activity against cancerous (Caco-2 and HeLa) cells than against normal (IEC-6) cells was observed. PFM were more inhibitory than CEs, and both generated oxidative stress in Caco-2 cells. PFM of L. plantarum 0991 and L. brevis 0983 induced apoptosis in Caco-2 cells by the mitochondrial signalling pathway. Conclusions: Anticancer activity of PFM and CEs of LAB, as well as the ability of apoptosis induction, is strain-specific.

Strain FM11-1 was isolated from flowers of Solanum torvum that were collected in Nakhon Si Thammarat Province, Thailand and was characterized using a polyphasic approach as Gram-positive, facultatively anaerobic with short chains and catalase-negative cocci. This strain produced L-lactic acid from glucose and was closely related to Enterococcus durans NBRC 100479T, Enterococcus faecium NRIC 1145T, Enterococcus hirae ATCC 9790T, Enterococcus lactis LMG 25958T and Enterococcus ratti DSM 15687T (98.92-99.73 %) based on 16S rRNA gene sequence similarity. The draft genome of strain FM11-1 had 2,784,928 bp and contained 2,586 coding sequences, with a genomic G+C content of 38.07 mol%. Values of ANIb, ANIm and digital DNA-DNA hybridization (dDDH) between strain FM11-1 and the closest strain, E. lactis LMG 25958T were 97.23%, 98.30% and 83.7%, respectively. The predominant cellular fatty acids were C19:0 cyclo 8c and C16:0. This strain was identified as Enterococcus lactis using polyphasic characterization and genome analysis. Strain FM11-1 contained genes involved in carbohydrate fermentation and specialty genes of antibiotic resistance. This strain showed adhesion ability (0.43±0.11) on Caco-2 cells but had no cytotoxicity against Caco-2, HepG2 and Vero cells.

Introduction: Probiotics are living microorganisms that when administered in appropriate amounts, provide several benefits such as prevention of infections, relief of allergic symptoms and lactose intolerance, control of serum cholesterol, blood pressure and gestational diabetes, reduction of gastrointestinal disorders, production of beneficial compounds such as vitamins, short chain fatty acids, and such microorganisms show evidence of actions against certain types of cancers. This study aimed to evaluate the evidence available in the literature on probiotics on breast cancer. Methods: It is an integrative review of the literature, through the PICO strategy, with the guiding question “Does probiotics consumption act on oxidative stress and reduces the risk of breast cancer”? The selection of articles was made on MEDLINE, SciELO and PubMed, including those published between 2016 and 2021, using the keywords: “breast cancer” AND “probiotics” AND “microbiota” their combinations in Portuguese and English, with with previously established inclusion criteria. Results and Discussion: The sample consisted of ten articles, and the answers found were: scientific research in vivo and in vitro, with some variations of probiotics strains, where all demonstrate actions and/or effects on cancer cells or acted on metabolic pathways mitigating the development of tumors. Conclusions: The manifested potential for a possible insertion in the treatment and prevention of breast cancer, being considered an innovation for the scientific community that still fights against various types of problems related to human health, especially some types of cancers, of negative prognoses and with difficult therapeutic interventions.
