Antiproliferation effects and antioxidant activity of two new Lactobacillus strains

Abstract: The microorganisms most commonly used as probiotics are lactic acid bacteria, especially those of the genus Lactobacillus. In the present study, two Lactobacillus strains, L. pentosus ITA23 and L. acidipiscis ITA44, previously isolated from mulberry silage, were characterized for their antiproliferative and antioxidant activities. The antiproliferative effects of the strains were investigated using the MTT assay with breast cancer (MDA-MB-231), liver cancer (HepG2) and normal liver (Chang) cell lines. The stra… Show more

“…The concentrations of the PFM are the most often given as a percentage or mg/mL, less often in µg/mL. The range of the tested concentrations is from 0.47% to 100% [ 11 , 35 , 36 , 37 , 38 , 39 ] or from 0.0001 to 1000 mg/mL [ 18 , 29 , 31 , 34 , 40 , 41 ]. In all of the above-mentioned studies, the MTT assay dominates as a tool of antiproliferative activity testing for cancerous cell lines and cytotoxicity assayed to normal cells.…”

“…Many research studies compared the antiproliferative action of PFM/CE/HK against cancerous vs. normal cell lines [ 11 , 17 , 18 , 29 , 35 , 37 , 40 , 45 , 46 ]. LAB lysates of eight different strains displayed variable inhibitory activity against cancerous vs. normal cell lines [ 46 ].…”

“…Cytotoxicity against Caco-2 (colon carcinoma, ATCC HTB-37), HepG2 (human hepatocyte carcinoma, ATCC HB-8065) and Vero (normal cells from African monkey kidney) cells were conducted by MTT assay (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) based on the activity of active mitochondrial enzymes [41,42]. In brief, cells were cultivated in MRS broth at 37°C for 48 h and then diluted in MRS broth (10 8 cells/ml).…”

“…The supernatant of the tested strain was determined for cytotoxicity against tumor cell lines including Caco-2 and HepG2 cells compared with Vero cells, as previously described [41,42]. Briefly, all cell lines were proliferated at 37°C in a humidified atmosphere containing 5% CO 2 in specific culture medium supplemented with 10% fetal bovine serum (FBS), penicillin (100 U/ml) and streptomycin (0.1 mg/ml).…”

“…The main beneficial effects on human health caused by probiotics are constantly described in the scientific literature, among them are: prevention of urogenital infections, immunomodulatory functions, control of serum cholesterol, blood pressure and gestational diabetes, relief of allergic symptoms and lactose intolerance, reduction of gastrointestinal disorders such as intestinal inflammatory syndromes, and prevention against certain types of cancer [2][3][4][5][6][7][8][9] .…”
