Effects of sciatic nerve transection on glucose uptake in the presence and absence of lactate in the frog dorsal root ganglia and spinal cord

Abstract: Frogs have been used as an alternative model to study pain mechanisms because the simplicity of their nervous tissue and the phylogenetic aspect of this question. One of these models is the sciatic nerve transection (SNT), which mimics the clinical symptoms of "phantom limb", a condition that arises in humans after amputation or transverse spinal lesions. In mammals, the SNT increases glucose metabolism in the central nervous system, and the lactate generated appears to serve as an energy source for nerve cell… Show more

“…an intact group) was not used because SNT did not induce significant changes in sham-operated frogs (Rigon et al, 2014;Burgess and McIlwain, 1994;Sinicropi et al, 1982). In addition, our AChE activity values showed a similar range as previously described for spinal cord of frog Rana pipiens (Sinicropi et al, 1982).…”

“…The SNT was made under anesthesia (prilocaine 0.1 mL/100 g body weight) and sterile conditions as described by Rigon et al (2014Rigon et al ( , 2013. For nerve transection the right sciatic nerve was randomly exposed and completely transected approximately 3 mm distal to the sciatic notch.…”

“…: IR-UU-AEC-3/1656/AD). Animals were anesthetized by intraperitoneal administration of ketamine hydrochloride 5%, 90 mg kg -1 (Ketaset 5%; Alfasan, Woerden, The Netherlands) and xylazine hydrochloride 2%, 5 mg kg -1 (Rompun 2%, Bayer, Leverkusen, Germany) and sterile conditions [27]. Surgery was performed in the control group without nerve transection, but in the other groups, complete left sciatic nerve transection was created.…”

“…The process of amphibian metamorphosis, characterized by behavioral, morphological, physiological and biochemical changes, is a classic model used to study the transition of vertebrates from an aquatic to a terrestrial environment (Bo et al, 2018;Gao et al, 2018;Kroth et al, 2018;Rigon et al, 2014).…”
