Parasite load, iNOS and cytokine profiles, and histopathological aspects of Leishmania infantum infection in dogs with different clinical presentations

Abstract: Visceral leishmaniasis (VL) is a zoonotic disease with a canine urban reservoir in South America. Dogs from an endemic area within Brazil, which were naturally infected with Leishmania infantum, and those presenting severe clinical (SC), mild, or no clinical (MNC) disease, were evaluated. Parasite load, histopathology, and cytokine and iNOS mRNA expressions were assessed in the spleen and liver in order to determine the potential markers for disease susceptibility or resistance. As a result, dogs with both S… Show more

“…The disease in dogs did not alter IL-6 levels in SL culture supernatants, as reported in previous studies [ 44 ]; however, high IL-6 mRNA expression was found in canine spleens [ 45 ]. Taken together, these results suggest post-transcriptional regulation of this cytokine in leishmaniasis, contributing for parasite replication in the host, because, interleukin 6 is a pro-inflammatory cytokine that participates in immune responses, parasite control, and tissue repair [ 46 ], therefore, their reduction can be one reason for disease susceptibility.…”

“…No correlation was found between the copy numbers recorded in ddPCR and the morphological changes in the spleen of the dogs included in the study. This finding is in concordance with the study of Vasconcelos et al, which demonstrated that dogs with severe disease, as well as asymptomatic or mildly symptomatic animals with moderate to extensive splenic tissue disorganization, can have high parasitic loads [44]. The authors suggested that ddPCR is advantageous over qPCR as it does not require a standard curve and is easy to standardize; therefore, it should be further used to investigate the relationship between parasitic load and the pathogenesis of the disease in dogs [43].…”
