Periodontitis is associated with reduced antioxidant capacity and increased oxidative damage. Oxidative stress induces inflammation and bone loss contributing to the pathological progression of periodontal disease. Calendula officinalis (CLO) has demonstrated anti-inflammatory and anti-oxidant activities. Therefore, the aim of this study was to evaluate the effect of CLO on oxidative stress and bone loss in rats subjected to experimental periodontitis (EP). For this, 72 male Wistar rats were divided into groups: Naïve, Saline (SAL) and CLO. Rats received SAL or CLO (90 mg/kg) 30 min before ligature and daily until the 11th day. Naïve group experienced no manipulation. After 11 days, the animals were euthanized and left maxillae collected for macroscopic analysis of alveolar bone loss (ABL). Periodontium was analyzed by macroscopy, scanning electron microscopy; confocal and light polarized microscopy. Immunohistochemical examination of DKK1, WNT 10b and β-catenin was performed. The gingival tissue was collected to reduced glutathione (GSH), superoxide dismutase (SOD), catalase (CAT) and malondialdehyde (MDA) analyses. The 11 days of ligature induced bone loss, breakdown of collagen fibers, increased the immunostaining DKK-1 while reduced WNT 10b and β-catenin expressions. Periodontitis reduced GSH, SOD, CAT and increase MDA. All findings were reversed by 90 mg/kg of CLO. In summary our findings demonstrated that CLO reduced oxidative stress and bone loss and preserved collagen fibers in rats with EP, with participation of WNT signaling pathway.

Bone loss caused by periodontitis can be potentiated by diabetes. Atorvastatin (ATV), has been tested in periodontitis due to its anti-inflammatory and antiresorptive activities, but it has been reported to increase the risk of diabetes. Therefore, this study aimed to evaluate the effect of ATV on alveolar bone of rats with periodontitis and diabetes. For this, 72 Wistar rats were divided into 4 groups: Naïve (N), experimental periodontitis (EP), diabetes mellitus (DM) and ATV. DM was induced by streptozotocin (60 mg/kg-ip.), after 12 hours fasting. Following, all animals were submitted to EP. ATV was administered (27 mg/kg, v.o.) 30 minutes before ligature and daily until the 11th day. Animals from N group did not undergo any intervention. After 11 days of EP, all animals had their glycemic rate measured and then were euthanized. Maxillae were collected for macroscopic, micro-tomographic and microscopic analyses. DM caused intense bone loss (60%) marked by reduction on trabecular thickness and bone volume (p<0.05). It was seen a significant reduction on osteoblast with increase on osteoclast counts, as well as, inflammatory infiltrate in the periodontium. ATV was not able to protect bone, improving bone loss by 21%. ATV caused a significant increase (20%) on blood glucose levels. In summary, ATV did not protect alveolar bone loss and did not modulate the inflammation in DM animals submitted to EP. ATV increase of blood glucose levels in these animals. Therefore, the systemic use of ATV in non-controlled diabetic conditions should be carefully evaluated.

Objectives:The aim of the study was evaluating the effect of adding simvastatin to synthetic bone substitute on the regenerative surgical treatment of bone defects associated with periimplantitis in a six months randomized controlled clinical trial.Material & Methods: 30 patients diagnosed with periimplantitis divided in two groups, group I treated by using synthetic bone substitute (Nanobone), group II treated by using Nanobone with simvastatin. Clinical evaluation was the measurements for probing pocket depth (PPD), clinical attachment level (CAL), plaque index (PI), modified sulcus bleeding index (mSBI) and mucosal recession. Radiographic bone fill was evaluated at baseline and after six months. Results:The results for both treatment groups showed significant reductions obtained in the mean PPD, CAL and mSBI at six months postoperatively when compared to baseline values, on the other-side no statistically significant difference was observed for the mean PI and MR. The mean of bon fill in group II was better than that of group I and this results was statistically significant (P < 0.05). Conclusions:The two regenerative approaches produced significant improvements in both clinical and radiographic assessments, but adding simvastatin to Nanobone produce better results in terms of bone fill.
