Cyrtopodium paludicolum germination with two Tulasnella isolates

Abstract: Symbiosis between orchid seeds and mycorrhizal fungi has been reported to be a determining factor in the success of germination and protocorm development in vitro. Th e aim of this study was to isolate and identify by molecular analysis the mycorrhizal fungus associated with Cyrtopodium paludicolum, and to evaluate its effi ciency in facilitating seed germination and development. Germination experiments were carried out using a fungus isolated from C. paludicolum (CH01) and Epidendrum secundum (M65), which has… Show more

“…In the germination of C. haagii (Table 1), the highest percentages of germinated seeds were obtained in the OMA medium combined with FHC3E (OMA + FHC3E), B&G + FHC, B&G + FHC3E and asymbiotic B&G, all these with germination values over 60 %. The values obtained for the best treatments are higher than those described by Carvalho et al (2018), who evaluated the percentage of in vitro symbiotic germination for the C. paludicolum orchid in association with mycorrhizal fungi, collected in the same geographic region, reaching 52.89 % of maximum germination.…”

“…According to Table 1 , many orchids are considered generalist—or anecdotally, promiscuous—forming OM with fungi isolated from another orchid species [ 6 , 48 , 56 , 57 , 58 , 59 , 60 ]. For specialists, there are no other options but to use OMF isolated from respective plants [ 61 ].…”

“…Some species are also very easy to germinate; they germinate in the absence of OMF, even when sown in a symbiotic medium. The rest of the germination assay, such as seed morphological changes, germination percentage, duration of germination and development of protocorms, and growth index, can be conducted like that of asymbiotic germination [ 14 , 26 , 34 , 51 , 60 , 69 ].…”

“…It could be noted that Brazil has the highest number of published works both on propagation and germplasm conservation, while for some countries only researches referring to propagation of native orchid species are reported and for other countries (Guyana, Paraguay, Trinidad and Tobago, Suriname, Uruguay) there were no registered works in any of the topics.
