Grape seeds have physical and physiological dormancy. Conventional germination methods are not very efficient. The in vitro culture of seeds and of mature zygotic embryos can be an alternative for overcoming germination barriers. The objective was to develop methodologies that increase seed germination and reduce the time to obtain seedlings. The following assays were developed: (1) conventional ex vitro germination, in a 2 × 3 factorial arrangement, of seeds with or without stratification for 90 days, from cultivars 'Niágara Rosada', 'Itália' and 'Red Globe'; (2) in vitro seed germination, in a 3 × 3 factorial arrangement, of the three cultivars and three explants (intact seeds, seeds sectioned in the micropyle region and seeds with a cross-section); and (3) in vitro germination of mature zygotic embryos, in a 4 × 2 factorial arrangement represented by four concentrations of mineral salts (MS, ½MS, WPM and ½WPM) and two cultivars ('Niágara Rosada' and 'Itália'). The conventional ex vitro germination assay resulted in a low seed germination for the three cultivars. The two types of section associated with in vitro culture provided an increase in seed germination. However, the highest germination rate (over 90%) in the shortest time (17 days) was achieved by the embryos grown in vitro. This study defines an efficient methodology to obtain high germination in grape seeds-over 85%-with uniformity and in a short time, using mature zygotic embryos grown in ½WPM culture medium. This methodology can accelerate grapevine breeding programs.

Intensive breeding programmes have been carried out to obtain high‐quality seedless grape cultivars. Seedless grape cultivars were used as pollinizer parents, but the number of hybrid seeds and the viability of the seed obtained could differ greatly. In this study, carried out over 3 years, the results obtained from crossing with some pollinizer cultivars belonging to different Vitis species were compared. Maternal cultivars that do not need emasculation (male sterile) and emasculated cultivars were also compared. Evaluations were based on the total number of hybrid seeds and the proportion of viable seeds. In addition, the effects of various treatments on the rates of germination of the hybrid seed were compared. Although the female parents (with female flower structure) produced more seed than did hermaphrodite mothers in 2019, different results were obtained in cross‐breeding studies in 2018 and 2020. Cold stratification was very effective in increasing germination by breaking dormancy of the hybrid seed. In addition, after treatment of cold moist storage (cold stratification), keeping the seed in distilled water overnight before planting increased germination. Although large numbers of seeds were produced by the genotype 86/1 and the cultivar ‘Beyaz Çavuş’, both of which do not require emasculation, many of these seeds did not germinate. Crosses with the cultivar ‘Red Globe’ as the maternal parent produced large numbers of viable seeds and high germination rates.

Giberellik asit (GA3) tüm bitkilerde olduğu gibi asmalar da endojen olarak sentezlenen, fizyolojik ve biyokimyasal birçok olayı etkileyen hayati bir hormon olmakla birlikte, bağcılıkta büyüme ve gelişimin düzenlenmesi amacıyla eksojen olarak en yaygın kullanılan bitki gelişim düzenleyicisidir. Islah çalışmalarında başlangıç materyali olarak kullanılan tohumun çimlenmesi ve fide haline dönüştürülmesi ıslah başarısının en önemli kriterleri arasında yer almakta olup, bu aşamada çeşitli nedenlerden dolayı kayıplar yaşanmaktadır. Bu çalışmada farklı konsantrasyonlardaki GA3 uygulamalarının melezleme ıslahı sonucunda elde edilen iki farklı Amerikan asma kombinasyonuna ait melez tohumların çimlenme özellikleri üzerine etkileri incelenmiştir. Türler arası melezlerin elde edilmesi amacıyla, ebeveyn olarak üç farklı asma türü (“Ramsey”, “Kober 5 BB” ve “Rupestris du Lot”) kullanılmış ve farklı konsantrasyonlardaki GA3 uygulamaları 2 farklı melez asma popülasyonu (“Ramsey × Rupestris du Lot” ve “Kober 5 BB × Rupestris du Lot”) üzerinde test edilerek, tohum çimlenmesi üzerine en etkili GA3 konsantrasyonunun belirlenmesine yönelik incelemeler gerçekleştirilmiştir. Araştırma bulgularına göre, “Ramsey × Rupestris du Lot” melezlerinde tohum çimlenme oranı 71,18 ile 77,64 arasında değişirken; “Kober 5 BB × Rupestris du Lot” melezlerinde 75,48 ile 77,40 aralığında değişim göstermiştir. Çimlenme oranı bakımından “Ramsey × Rupestris du Lot” melezleri için optimal uygulama konsantrasyonunun, 500-2000 ppm GA3 olduğu kaydedilirken; “Kober 5 BB × Rupestris du Lot” melezleri için en etkili dozun 500-1000 ppm GA3 olduğu belirlenmiştir. Bu çalışmanın, melezlemeyle elde edilen asma tohumlarının çimlenme oranlarını arttırmak amacıyla yürütülecek diğer araştırmaları kolaylaştıracağı ve ayrıca asma germplazmının daha etkin kullanılması amacıyla düşük tohum çimlenme oranı sorununun ele alınması bakımından da önemli bir referans sağlayacağı düşünülmektedir.
