Red blood cells derived from peripheral blood and bone marrow CD34+ human haematopoietic stem cells are permissive to Plasmodium parasites infection

Abstract: The production of fully functional human red cells in vitro from haematopoietic stem cells (hHSCs) has been successfully achieved. Recently, the use of hHSCs from cord blood represented a major improvement to develop the continuous culture system for Plasmodium vivax. Here, we demonstrated that CD34+hHSCs from peripheral blood and bone marrow can be expanded and differentiated to reticulocytes using a novel stromal cell. Moreover, these reticulocytes and mature red blood cells express surface markers for entra… Show more

“…JkRBCs Support Invasion by P. falciparum. We next tested the ability of jkRBCs to support P. falciparum invasion, as has been observed for other nucleated erythroid precursors (14,30,58). Indeed, we observed invasion into jkRBCs by two different strains of P. falciparum: the sialic acid-independent strain 3D7 (59) and the sialic acid-dependent strain Dd2 (Fig.…”

“…In addition, it is possible to culture hematopoietic stem/progenitor cells (HSPC)/CD34 + cells to induce erythroid differentiation and consequently produce reticulocytes in vitro [8] . Reticulocytes generated from CD34 + cells from both bone marrow (BM) and peripheral blood mononuclear cells (PBMC) have been previously used for culturing P. vivax ; but the authors did not provide data regarding the efficiency of invasion and the development of the parasites in vitro [9] .…”

“…Potential that bone marrow could be the source for reticulocytes for in vitro cell culture is becoming an increasingly important area of P. vivax research. Fernandez-Becerra et al have expanded CD34 pos hematopoietic stem cells to generate CD71 pos reticulocytes for in vitro P. vivax culture (63). Others have concentrated their efforts on enriching reticulocytes from cord blood.…”
