Leishmania amazonensis DNA in wild females ofLutzomyia cruzi (Diptera: Psychodidae) in the state of Mato Grosso do Sul, Brazil

Abstract: Studies on natural infection by Leishmania spp of sandflies collected in endemic and nonendemic areas can provide important information on the distribution and intensity of the transmission of these parasites. This study sought to investigate the natural infection by Leishmaniain wild female sandflies. The specimens were caught in the city of Corumbá, state of Mato Grosso do Sul (Brazil) between October 2012-March 2014, and dissected to investigate flagellates and/or submitted to molecular analysis to detect L… Show more

“…An ideal DNA extraction protocol should meet some criteria such as concentrated yield, elimination of PCR inhibitors, implementable as laboratory technique, low cost, reproducibility, and safety (Rantakokko-Javala and Javala, 2002). Researchers have used different extraction methods to extract insect DNA, such as phenolchloroform-isoamyl alcohol, detergents like Triton X-100 and Triton X-100/DDT, silica column solid phase extraction, and Chelex ® Molecular Biology Grade Resin (Bio-Rad Laboratories, Hercules, USA) (Cova et al, 2015;Oliveira et al, 2015;Paiva et al, 2007;Pinto et al, 2015;Santos et al, 2013).…”

“…The contents of the slide and the digestive tube were ground using a plastic pestle in 1.5-mL tubes with 300 μL of 5% Chelex resin solution (Bio-Rad, Hercules, CA, USA). DNA extraction was performed according to the protocol described by Oliveira [ 28 ].…”

“…Thereafter, collected females that did not have blood in their gut were dissected, and identified on the basis of their spermatheca characteristics. These females were then placed in 1.5-mL microtubes with isopropyl alcohol to determine the presence of Leishmania DNA, the results of which are described by Oliveira et al [21]. …”
