Bioassay guided purification of cytotoxic natural products from a red alga Dichotomaria obtusata

Abstract: tDifferent solvent extracts of Dichotomaria obtusata (J. Ellis & Solander) Lamark, Galaxauraceae, a redalgae collected from the coast of Bushehr in the Persain Gulf, was investigated for its cytotoxic prop-erties and chemical constituents. The fresh alga, after extraction with methanol and dichloromethanewere combined and partitioned between water, dichloromethane and ethyl acetate. The above fractionswere then tested against MOLT-4 (human lymphoblastic leukemia) cancer cell line. The IC50values ofthe dichloro… Show more

“…The test exhibited that six isolates produced the extract that showed activity against one pathogen, and two isolates produced extract that was active against two pathogens (Table 3.). Methanol and ethyl acetate were used for extraction to provide the polar and less-polar compounds, respectively (Jassbi et al 2016).…”

“…Additionally, 37 compounds, including mostly fatty acids with some diols, alkenes, alcohols, amides and aldehydes, were extracted from the alga Halymenia durvillei Bory de Saint-Vincent, identifying by GC-MS analysis (Tassakka et antibacterial and antioxidant steroids and fatty acids from a red alga, Hypnea agelliformis Greville ex J.Agardh, and two brown algae, Cystoseira myrica (S.G.Gmelin) C.Agardh, and Sargassum boveanum J.Agardh. In continuing our research on the Iranian marine algae, the bioassay guided puri cation of a red algae Dichotomaria obtusata (J.Ellis & Solander) Lamarck, yielded two terpenoid derivatives trans-phytyl palmitate and γ-tochopherol that showed signi cant cytotoxicity against three human cancer cell lines (Jassbi et al 2016). Recently, Moheimanian et al (2021) investigated total phenolic content, antibacterial, cytotoxic and antioxidant activities of ve brown algae from the Persian Gulf including Colpomenia sinuosa (Mertens ex Roth) Derbès & Solier, Iyengaria stellate (Børgesen) Børgesen, Padina gymnospora, (Kützing) Sonder, Sargassum acinaciforme Montagne, and Sirophysalis trinodis (Forsskal) Kützing.…”

“…Top-down approaches towards the discovery of NPs have traditionally been the primary means of NP discovery, and, to date, this is still a successful strategy. However, genome-guided strategies are being incorporated as alternative or parallel approaches to NP discovery, as they allow for the investigation of the biosynthetic potential and metabolic capacity of microorganisms rather than just what can be detected in assays [ 7 , 8 , 9 , 10 ]. In addition, by combining integrative omics and bioinformatic analysis, insight into the activation of BGCs that are silent under standard laboratory culturing conditions could be provided.…”
