Background: Mahasudarshan Churna (MC) is a polyherbal Ayurvedic medicine that is employed in fever (especially chronic type), cold and malaria, improvement of digestion and appetite, removes toxins from the blood, boosts immunity and protects against common bacterial infections. Methods: Validation and quantification of oleanolic acid (OA), ursolic acid (UA), mangiferin (M), gallic acid (GA), quercetin (Q) and curcumin (C) in commercial MC formulations by HPTLC method. Mobile phase, hexane: ethyl acetate: acetone (16.4: 3.6: 0.2, v/v) was used for the separation of OA and UA; ethyl acetate: glacial acetic acid: formic acid: water (20: 2.2: 2.2: 5.2 v/v) for the development of M; and toluene: ethyl acetate: formic acid (13.5: 9: 0.6 v/v) for the separation of GA, Q and C in crude sample extracts. Visualization and scanning were performed at λ = 530 nm for OA and UA, at λ = 254 nm for M and at λ = 366 nm for GA, Q and C. In addition, HPLC-PDA analysis was used to confirm the HPTLC results. Results: Major bio-active compounds in MC formulations were oleanolic acid (1.54-1.78%), mangiferin (1.38-1.52%) and gallic acid (1.01-1.15%); followed by ursolic acid (0.79-0.98%), curcumin (0.45-0.67%) and quercetin (0.22-0.34%).

The rapid, highly-sensitive and ecologically greener reversed-phase (RP)/normal-phase (NP) high-performance thin-layer chromatography (HPTLC) densitometric technique has been developed and validated for the determination of trans-resveratrol (TRV). The reversed-phase HPTLC-based analysis of TRV was performed using ethanol–water (65:35, v v−1) combination as the greener mobile phase, while, the normal-phase HPTLC-based estimation of TRV was performed using chloroform–methanol (85:15, v v−1) combination as the routine mobile phase. The TRV detection was carried out at 302 nm for RP/NP densitometric assay. The linearity was recorded as 10–1200 and 30–400 ng band−1 for RP and NP HPTLC techniques, respectively. The RP densitometric assay was observed as highly-sensitive, accurate, precise and robust for TRV detection in comparison with the NP densitometric assay. The contents of TRV in commercial formulation were recorded as 101.21% utilizing the RP densitometric assay, while, the contents of TRV in commercial formulation were found to be 91.64% utilizing the NP densitometric assay. The greener profile of RP/NP technique was obtained using the analytical GREEnness (AGREE) approach. The AGREE scales for RP and NP densitometric assays were estimated 0.75 and 0.48, respectively. The recorded AGREE scale for the RP densitometric assay indicated that this technique was highly green/the ecologically greener compared to the NP densitometric assay. After successful optimization of analytical conditions, validation parameters, AGREE scale and chromatography performance, the RP densitometric assay with univariate calibration was found to be better than the NP densitometric assay for the analysis of TRV.

Das et al.: Analytical Methods of Standardization of Asava and Arista Plants serve as a rich source of bioactive molecules, which are used to treat various diseases in Ayurveda. Medicinal plant materials are formulated into valuable Ayurvedic medicines by application of modern scientifi c techniques, where standardization plays a pivotal role for authentication. Standardization confi rms the identity, quality and purity of drugs. World Health Organization has set up suitable specifi c standardization parameters to evaluate the crude drugs and their fi nished products. These include various evaluation techniques such as pharmacognostical, physico-chemical, phytochemical, analytical, biological and biotechnological. Nowadays, application of several modern analytical techniques have become inevitable for evaluating the polyherbal Ayurvedic formulations to ensure quality, safety and effi cacy. Various spectroscopic and chromatographic methods applicable for this purpose are ultraviolet spectroscopy, Fourier-transform infrared spectroscopy, nuclear magnetic resonance spectroscopy, thin-layer chromatography, high performance thin layer chromatography, high performance liquid chromatography, gas chromatography, mass spectroscopy and hyphenated techniques such as gas chromatography-mass spectroscopy, liquid chromatography-mass spectroscopy, liquid chromatography-nuclear magnetic resonance spectroscopy. Out of several formulations available in Ayurveda, asavas and aristas are considered as unique dosage forms due to their indefi nite shelf life. Standardization of different asava-arista formulations using various analytical techniques are vividly discussed in this review.
