1α,25‐dihydroxyvitamin D3 acts via transforming growth factor‐β to up‐regulate expression of immunosuppressive CD73 on human CD4+ Foxp3 T cells

Abstract: SummaryVitamin D deficiency is associated with increased incidence and severity of various immune‐mediated diseases. Active vitamin D (1α,25‐dihydroxyvitamin D3; 1,25(OH)2D3) up‐regulates CD4+ T‐cell expression of the purine ectonucleotidase CD39, a molecule that is associated with the generation of anti‐inflammatory adenosine. Here we aimed to investigate the direct impact of 1,25(OH)2D3 on expression of the downstream ecto‐5′‐nucleotidase CD73 by human CD4 T cells, and components of the transforming growth f… Show more

“…The negative correlation identified between serum levels of TGF-β-1 and serum vitamin D levels (p < 0.01) highlights other studies in MS and findings that vitamin D supplementation may increase TGF-β expression, which may play a role in its mechanism of action [35][36][37]. TGF-β-1 is an anti-inflammatory cytokine produced by regulatory T-cells.…”

“…Since there was no evidence of PD1 expression (i.e., antigen experience) after nonliposomal delivery, these data suggest direct effects of calcitriol on the TCR-transgenic T cells. Direct T cell effects of vitamin D3 were noted previously, due to enhanced TGF-β bioactivity (40). Furthermore, differentiation of antigen-specific effector Tregs and development of anergy require antigen experience.…”

“…Moreover, in utero vitamin D deficiency in mice leads to increased airway smooth muscle mass and airway resistance [25]. Furthermore, in children with severe asthma, lower levels of vitamin D have been shown to be associated with increased airway smooth muscle mass [26, 27]. Recently, 1,25(OH)2D3 has been reported to exert a negative regulatory effect on VEGF secretion [5, 28].…”
