Analytical chemistry (I) is one of the core courses in chemistry major. We have performed the practical exploration, in which the scientific research achievements are integrated into the teaching contents and the latter are used to activate the research inspiration, in order to improve the teaching quality, enhance the students' interest in learning, and realize the mutual promotion between teaching and scientific research. The actual results are satisfactory.
The optimal conditions for extracting procyanidins from Lycium ruthenicum Murr. with sub-critical fluid R134a (1,1,1,2-tetrafluoroethane) in a 1 L extraction vessel were studied. Taking the extraction ratio of procyanidins as an indicator, the influences of pressure, temperature, and extraction time on extraction rate of procyanidins from Lycium ruthenicum Murr. were studied by single factor experimental methods and orthogonal array design. The order of factors affecting extraction ratio of procyanidins was extraction temperature > extraction pressure > extraction time. The optimum extraction conditions were as follows: the extraction ratio of procyanidins from Lycium ruthenicum Murr. was the highest with a extraction pressure of 1.2 MPa, extraction temperature of 50˚C and extraction time of 90 min. While the contents of procyanidins in Lycium ruthenicum Murr. from different origins were determined by vanillin-hydrochloric acid method under the optimal conditions. The method has the advantages of easy operation, good selectivity, low extraction temperature and high extraction efficiency, which is suitable for extraction of procyanidins components in Lycium ruthenicum Murr.
