Мета. Визначити проблеми збереження видів Carlina аcaulis L., C. cirsioides Klok та C. onopordifolia Besser ex Szafer, Kulcz. et Pawl. in situ та ex situ та оптимізувати умови їх введення in vitro. Методи. Для отримання та вкорінення асептичних проростків насіння піддавали передпосівній обробці розчином гіберелової кислоти (ГК3) або індолілмасляної кислоти (ІМК). Простерилізоване насіння висаджували на агаризоване живильне середовище Мурасіге-Скуга з половинним вмістом макро- і мікросолей (МС/2), без регуляторів росту. Для мікроклонального розмноження цих видів використовували розетки 2–3 місячних особин, які культивували на агаризованому середовищі МС/2, доповненеому кінетином (Кін) (від 1–3 мг/л) та 0,1 мг/л 1-нафтилоцтової кислоти (НОК). Результати. З’ясовано, що при замочуванні насіння С. acaulis, С. cirsioides та С. onopordifolia у розчині ГК3, відсоток формування коренів складав 33,3 %, 33,3 % та 22,2 % відповідно. Передпосівна обробка насіння перед стерилізацією у розчині ІМК концентрацією 1000 мг/л протягом 2–4 год була ефективніша. Відсоток формування коренів у C. аcaulis, C. cirsioides і C. onoрordifolia був у 2,4–4,5 рази вищий у порівнянні з обробкою розчином ГК3. Середовище МС/2, доповнене НОК та Кін, найкраще забезпечувало формування мікроклонів. У рослин C. cirsioides цей показник за 6 місяців культивування становив 6,6–6,8 розеток на живець, у рослин С. acaulis та C. onopordіfolia — 4,2–5,0 та 4,8–5,2 відповідно. Для підвищення відсотку вкорінення мікроклонів доцільним виявилось замочування їх у розчині ІМК концентрацією 1000 мг/л упродовж 1 хв. Висновки. Досягнено підвищення відсотків укорінення рослин C. сirsioides і C. оnoрordifolia до 100 %, C. аcaulis — до 80 %, а також уникнено травмування проростків і змін концентрації розчину ІМК, які можуть виникати під час стерилізації за високих температур, унаслідок використання нестерильного розчину цього регулятора росту. Підібрано умови для мікроклонального розмноження С. acaulis, C. cirsioides і C. onopordіfolia та розроблено схеми вкорінення отриманих мікроклонів in vitro.
