В настоящее время показано, что нарушение нормального функционирования клеток церебрального эндотелия способно приводить к различным патологиям, в том числе и к развитию нейродегенеративных заболеваний. Доказано, что коэнзим Q10 обладает нейропротекторным действием и является перспективным препаратом для лечения и профилактики ряда заболеваний, вызванных митохондриальными нарушениями. Изучение действия коэнзима Q10 на клетки церебрального эндотелия является важным и интересным направлением в нейрофармакологии, открывающим новые перспективы для решения вопроса управления проницаемостью ГЭБ, предотвращения развития нейровоспаления и нейродегенерации.
Оценивали способность убихинола, введенного внутривенно, влиять на содержание и редокс-статус CoQ10 (отношение концентрации убихинола к общей концентрации CoQ10) в тканях крысы. Содержание убихинола и общего пула CoQ10 до и в течение 8 сут после внутривенного введения 1 % водного раствора солюбилизированной субстанции убихинола в дозе 30 мг/кг определяли методом ВЭЖХ с электрохимическим детектированием. Впервые показано, что, независимо от изменений концентрации CoQ10 в органах в результате внутривенного введения убихинола, редокс-статус в миокарде и головном мозге остается неизменным, а в печени постепенно возрастает. В плазме крови доля убихинола в общем содержании CoQ10 постоянна на протяжении, как минимум, 2 сут после введения и максимальна среди всех изученных тканей. Различия редокс-статуса CoQ10 в плазме и органах свидетельствуют о частичном окислении убихинола при поступлении из крови в органы до уровня их эндогенного тканевого редокс-баланса.
