Objective. Emerging data indicate that oxidative stress is closely associated with the pathogenesis of cardiovascular disease in type 2 diabetes mellitus (T2DM). The present study aimed to assess the effect of the most abundant flavonoid in the human diet quercetin (Q) on the myocardial redox status in rats with T2DM. Methods. T2DM was induced in male Wistar rats by a high caloric diet (for 14 weeks) and two streptozotocin (25 mg/kg b.w.) injections applied in four weeks of the diet, once a week for two weeks. The Q was administered intragastrically by gavage in a dose of 10 or 50 mg/kg of the body weight for 8 weeks starting from the 8th day after the last streptozotocin injection. The control rats received citrate buffer and seven days after the last STZ injection, basal glucose levels were measured in all animals. Results. Administration of Q increased insulin sensitivity in diabetic rats with more pronounced effect at a dose of 50 mg/kg b.w. The Q also decreased free radical oxidation in the heart mitochondria of diabetic animals, thus limiting the formation of advanced oxidation protein products in a dose-dependent manner and normalized the activity of antioxidant enzymes (superoxide dismutase, glutathione peroxidase, glutathione reductase) in cardiac mitochondria independently of the dose used. In addition, the Q in both doses prevented the development of oxidative stress in the T2DM rats cardiomyocytes by reducing NADPH oxidase and xanthine oxidase activities. Conclusions. The findings demonstrate that Q in both doses 10 mg/kg and 50 mg/kg can protect from the development of oxidative stress in cardiomyocytes in the diabetic rats. The present data indicate that the use of Q may contribute to the amelioration of cardiovascular risk in patients with T2DM.

Індукцію МС розпочинали через 2 тижні після хірургічного втручання шляхом утримання тварин на високовуглеводній дієті, яка забезпечувалася вільним доступом щурів до 30% розчину сахарози в якості питної води протягом 5 тижнів. Встановле-но, що гіпоестрогенія призвела до зниження рівня лептину. При поєднанні дефіциту естрогенів із МС рівень лептину залишався таким же низьким, як і у оваріектомованих щурів без МС. Менопауза характеризується зміненим гормональним статусом, в першу чергу зниженням рівня естрогенів. Саме з прогресуючою втратою естрогенів пов'язано підвищення ризику розвитку у жінок в постменопаузі ожиріння, діабету і серцево-судинних захворювань. Результати епідеміологіч-них досліджень свідчать, що захворювання серце-во-судинної системи є відповідальними за третину усіх смертей жінок у світі та половину усіх смертей жінок у віці понад 50 років [1]. Головним фактором ризику серцево-судинної патології у жінок цієї віко-вої категорії вважають метаболічний синдром (МС), незалежним предиктором компонентів якого визнано менопаузу [2].Одним із гормонів/адипокінів, що секретує жи-рова тканина, є лептин, який відіграє важливу роль в центральному контролі енергетичного метаболіз-му, а також володіє багатьма плейотропними ефектами в різних фізіологічних системах [3]. При дослідженні ролі лептину в метаболічних процесах виявлено тісний зв'язок із інсулінорезистентністю та абдомінальним ожирінням, основними складо-вими МС. Показано, що інсулін може регулювати синтез лептину адипоцитами, а хронічна гіперінсу-лінемія, яка є характерною для постменопаузаль-ного МС, збільшує секрецію лептину [4].Одна із периферичних функцій лептину поля-гає у взаємодії між енергетичним обміном і імун-ною системою, тому йому відводять важливу роль у новій області -імунометаболічній [5]. Лептинові рецептори експресуються на усіх клітинах імунної системи, а регуляторні ефекти лептину включають вплив на фактори як вродженої, так і адаптивної імунної відповіді. Експериментальна медицина і морфологіяУкраїнський журнал медицини, біології та спорту -№ 5 (7) 33 В статті наведені результати експерименталь-ного дослідження стану метаболічної ендотоксемії за умов дефіциту естрогенів у поєднанні з метабо-лічним синдромом та застосування сполук з імуно-модулючими та естрогенними властивостями. Встановлено, що гіпоестрогенія є самостійним чин-ником підвищення рівня антитіл до ендотоксину в оваріоектомованих щурів. Поєднання дефіциту естрогенів з метаболічним синдромом додатково не впливає на цей показник. Сполуки з імуномоду-люючою активністю левамізол та його похідне ПЛ-308 нормалізують рівень ендотоксину в оваріо-ектомованих щурів з метаболічним синдромом, тоді як введення 17β-естрадіолу знижує, проте не нормалізує рівень антитіл до ліпополісахариду. Серцево-судинні захворювання є основ-ною причиною смертності у жінок в постменопаузі. За результатами Framingham Study (Stangl V, 2002) продемонстровано, що щорічна серцево-судинна захворюваність у жінок у віці до 55 років в премено-паузі в порівнянні з жінками в тій же віковій групі, в якій мала місце менопауза, була на 50% нижче.Одним із факторів, які впливають на підвищен-ня захворюваності у жінок в постменопаузі є мета-болічний синдромом (МС). Ряд досліджень показу-ють більш високу наявність МС у жінок після мено-паузи, в порівнянні з пременопаузою [11]. Пошире-ність МС у жінок після менопаузи складає від 22% до 69%, змінюючись від однієї країни до іншої, а також в залежності від використовуваної методо-логії [16]. Показано також, що МС пов'язаний із підвищеним ризиком смертності жінок в постмено-паузі, ніж для жінок в пременопаузі [15].Не лише підвищення кардіоваскулярної патоло-гії характерне для постменопаузи, а й діабет 2 типу, аутоімунні захворювання, неоплазії тощо [9].Важливим патогенетичним чинником МС вва-жають метаболічний дисбаланс, що супроводжу-ється ожирінням, резистентністю до інсуліну, та підвищення маркерів/медіаторів запалення.Вже давно в клінічній практиці, особливо хірур-гічній, для оцінки загального стану пацієнтів засто-совують показники, які характеризують синдром ендогенної інтоксикації [14].
