The porcine reproductive and respiratory syndrome (PRRS) caused the serious economic damage to swine breeding around the world. It is a viral infective disease against which live attenuated and inactivated vaccines are not always successful. Development of new types of drugs such as DNA vaccines is necessary for improving the protection against the virus. DNA vaccines induce the development of both a cellular and humoral immune response. Such vaccines consist of a plasmid or viral vector with genes of potentially immunogenic proteins. The expression of these genes realized in cells of the vaccinated animal. It leads to the synthesis of antigen proteins triggering the immune response. The purpose of this work is to create a genetic construction that can be used as DNA vaccine against PRRS virus. The construction consists of the commercial vector pVAX1 and open reading frame of two structural proteins of PRRS virus, a lysosomal localization signal sequence of the invariant chain gene and regulatory elements necessary for the expression of cloned genes in mammalian cells.
Aims To study indices of the cytokine state in umbilical blood of newborns born by women with chronic inflammatory gynaecological diseases in case of a late manifestation of the infectious process. Methods 37 newborns born by women with chronic inflammatory gynaecological diseases were examined. The levels of interleukin-2 (IL-2), tumour necrosis factor (TNF-α), interferon gamma (IFN-γ) in umbilical blood were analysed using a kit for the enzyme-linked immunosorbent assay ProConIL-2, IFNgamma, TNF-α (“Protein Contour” Limited Liability Company, St. Petersburg). The following software packages were used: Statistica version 6.0 and EXCEL 2003, SPSS 13.0, MegaStat. PolyAnalist 3.5 Pro package was used for the analysis of multidimensional nonlinear dependences. Results Manifestations of the infectious process were not observed in all newborns within the first month of their life. From the end of the first month and up to the third month of life, clinical presentations of the infection, including viral infection, were observed in 15 newborns, cytomegalovirus DNA was detected in urine analyses that allowed diagnosing cytomegalovirus infection. In 22 newborns no clinical presentations of the infection were recorded. The control group was comprised of 15 newborns without infection. Analysis of multidimensional nonlinear dependences using “PolyAnalist 3.5. Pro” package has shown factors which are significant for prognosis of the infectious process in the postnatal period: IL-2, TNF-α, IFN-γ. Formula of dependence of the infectious process prognosis on the content of IL-2, TNF-α, IFN-γ in umbilical blood of newborns born by mothers with chronic inflammatory gynaecological diseases in case of a late detection of cytomegalovirus DNA was suggested: IL-2* TNF-α <–527.79 + 4.1342* IFN-γ * TNF-α. If this inequality is fulfilled, we prognosticate an infectious process in the postnatal period. Accuracy is 85%. If this inequality is not fulfilled, the infection will not develop. Accuracy is 91%. P < 0,000001. Sensitivity is 87%, specificity is 91%. Positive predictive value is 9.6; negative predictive value is 7. Conclusions The formula is an early marker of the infectious process manifestation in newborns born by mothers with chronic inflammatory gynaecological diseases and it allows separating children of the risk group for the appropriate therapy prescription.
Изучить аллергические проявления у детей с минимальной мозговой дисфункцией и особенности их лечения. Материалы и методы. В исследование включены 82 ребенка с аллергическими проявлениями на фоне минимальной мозговой дисфункции. Результаты. В статье представлены данные о патогенезе аллергических проявлений у детей с минимальной мозговой дисфункцией. Авторы излагают собственный опыт использования гидроксизина, обладающего анксиолитическим и антигистаминным действием, у детей с острыми аллергическими реакциями на фоне минимальной мозговой дисфункции. Заключение. Опыт применения гидроксизина показал высокую эффективность препарата и безопасность его применения у детей для купирования аллергичеcких и невротических проявлений.
Цель. Определить клинико-морфологическую и микробиологическую эффективность лечения язвенной болезни желудка, ассоциированной с Helicobacter pylori, препаратом рекомбинантного IL-2-«Ронколейкин» без применения антибактериальных препаратов. Материалы и методы. 108 человек, страдающих ЯБЖ, ассоциированной с НР, были рандомизированно разделены на две группы. Пациенты II группы получали стандартную трех-или четырехкомпонентную терапию. Больные, включенные в I-основную группу, находились на аналогичном лечении, но вместо антибиотиков им через гастроскоп по периферии язвенного дефекта в 4-6 точек в подслизистую вводили 0,1 мг рекомбинантного IL-2-Ронколейкина (Биотекс, Россия), одновременно 0,4 мг того же препарата вводили в/в. Процедуру проводили трижды с интервалом 72 часа. Результаты. Через 1 месяц после окончания терапии отмечено, что в I группе эрадикация НР составила 95,4% против 81,5% в контроле. В основной группе средний период эпителизации язвы составил 10,79±0,46 дней против 35,23±1,58 у пациентов, получавших традиционную терапию. В третий месяц мониторинга в биоптате слизистой оболочки желудка у больных основной группы в 93,5% случаев нет морфологических признаков воспаления и у 81,4% больных нет кишечной метаплазии. Заключение. Предлагаемый метод комплексного лечения язвенной болезни желудка, ассоциированной с Helicobacter pylori, является эффективным, клинически и патогенетически обоснованным.
