Целью нашего исследования явилось изучение распределения частот аллелей полиморфизма rs9939609 гена FTO, полиморфизма rs738409 гена PNPLA3 и полиморфизма rs58542926 гена TM6SF2 среди якутской популяции. Протестировано всего 86 образцов ДНК здоровых добровольцев якутской национальности. Анализ распределения частот аллелей и генотипов полиморфного варианта гена FTO (rs9939609) в исследованной группе не выявил достоверных отличий. Мутантная аллель А наиболее чаще встречалась в выборке женщин. В выборке женщин, распределение частот аллелей и генотипов выявил достоверное (р <0,01) преобладание генотипа TT (13%) и аллели T (26%). Анализ распределения частот аллелей и генотипов полиморфного варианта гена PNPLA3 (rs738409) не выявил достоверных отличий, в обоих группах преобладал аллель G и гомозиготный генотип GG. Результаты анализа распределение частот аллелей и генотипов полиморфизма rs58542926 гена TM6SF2 в группе мужчин и женщин выявил преобладание лиц с генотипом СС (89 % и 90 % соответственно). The aim of our study was to study the frequency distribution of alleles of the rs9939609 polymorphism of the FTO gene, the rs738409 polymorphism of the PNPLA3 gene, and the rs58542926 polymorphism of the TM6SF2 gene among the Yakut population. A total of 86 DNA samples of healthy volunteers of Yakut nationality were tested. Analysis of the frequency distribution of alleles and genotypes of the polymorphic variant of the FTO gene (rs9939609) in the study group did not reveal significant differences. Mutant allele A was most common in the sample of women. In the sample of women, the distribution of allele and genotype frequencies revealed a significant (p <0.01) prevalence of the TT genotype (13%) and the T allele (26%). Analysis of the frequency distribution of alleles and genotypes of the polymorphic variant of the PNPLA3 gene (rs738409) did not reveal significant differences; the G allele and the homozygous GG genotype prevailed in both groups. The results of the analysis of the frequency distribution of alleles and genotypes of the rs58542926 polymorphism of the TM6SF2 gene in the group of men and women revealed the prevalence of persons with the CC genotype (89% and 90%, respectively).
Цель исследования – поиск ассоциаций полиморфизмов генов LPS, ABCB1 и MTHFR на риск возникновения врожденных расщелин губы и нёба. Выборка исследованных лиц состояла из 94 детей с врожденными расщелинами. Группой сравнения служила, выборка здоровых добровольцев в количестве 156 человек. Анализ распределения частот аллелей и генотипов в выборке детей с лицевыми расщелинами и здоровой выборкой, показал достоверное (р = 0,000) различие генотипов только по полиморфизму rs1045642 гена ABCB1. Расчет отношения шансов показал достоверно высокий риск развития врожденных лицевых расщелин у гомозиготных носителей аллели Т (OR = 2,705 при 95% CI = 1,448–5,052). При анализе частот аллелей и генотипов в зависимости от тяжести расщелин, в группах врожденная расщелина губы (р = 0,000) и врожденная расщелина губы и нёба (р = 0,006) достоверное различие только по генотипу ТТ полиморфизма rs1045642 гена ABCB1. При этом носительство гомозиготного генотипа ТТ предполагает высокому риску и врожденной расщелины губы (OR = 3,114; 95% CI = 1,123–8,634) и врожденной расщелины губы и нёба (OR = 2,804; 95% CI = 1,333–5,895). При анализе частот аллелей и генотипов в выборке детей с расщелиной неба и здоровой выборки, достоверную (р = 0,035) значимую разницу показали гомозиготные носители аллеля Т по полиморфизму rs2235371 гена LPS. Носительство генотипа ТТ показал достоверно высокий риск развития врожденной расщелины нёба у гомозиготных носителей аллели Т (OR = 5,429 при 95% CI = 1,135–25,962). Таким образом исследование достоверно высокий риск возникновения врожденных лицевых расщелин был установлен у носителей генотипа ТТ полиморфизма rs1045642 гена ABCB1, при этом носительство гомозиготного генотипа ТТ по полиморфизму rs2235371 гена LPS достоверно предрасполагала к риску возникновения врожденной расщелины нёба. The aim of the study was to fi nd associations of polymorphisms of the LPS, ABCB1 and MTHFR genes for the risk of congenital cleft lip and palate. The sample of studied individuals consisted of 94 children with congenital cleft. The comparison group was a sample of 156 healthy volunteers. Analysis of the distributionof allele and genotype frequencies in a sample of children with facial cleft and a healthy sample showed a signifi cant (p = 0.000) difference in genotypes only by the rs1045642 polymorphism of the ABCB1 gene. The calculation of the odds ratio showed a signifi cantly high risk of developing congenital facial cleft in homozygous carriers of the T allele (OR = 2.705 at 95% CI = 1.448–5.052). When analyzing the frequency of alleles and genotypes depending on the severity of cleft, in the groups congenital cleft lip (p = 0.000) and congenital cleft lip and palate (p = 0.006), the only signifi cant difference was in the genotype of the TT polymorphism rs1045642 of the ABCB1 gene. At the same time, carrying the homozygous TT genotype implies a high risk of both congenital cleft lip (OR = 3,114; 95% CI = 1,123–8,634) and congenital cleft lip and palate (OR = 2,804; 95% CI = 1,333–5,895). When analyzing the frequencies of alleles and genotypes in a sample of children with cleft palate and a healthy sample, a signifi cant difference (p = 0.035) was shown by homozygous carriers of the T allele according to the rs2235371 polymorphism of the LPS gene. Carrying the TT genotype showed a signifi cantly high risk of developing congenital cleft palate in homozygous carriers of the T allele (OR = 5.429 at 95% CI = 1.135–25.962). Thus, a signifi cantly high risk of congenital facial cleft was found in carriers of the TT genotype of the rs1045642 ABCB1 gene polymorphism, while the carrier of the homozygous TT genotype by the rs2235371 polymorphism of the LPS gene signifi cantly predisposed to the risk of congenital cleft palate.
