На модели ишемия-подобного состояния в смешанной нейроглиальной культуре новорожденных мышей показано, что новое производное 3-гидроксипиридина с остатком аскорбиновой кислоты (соединение 3-ЕА) уменьшает некротическую гибель клеток коры головного мозга (нейроны и астроциты) в среднем на 28 %, p ≤ 0,05 при ишемии и ишемии/реоксигенации (после OGD/R) in vitro за счет дозозависимого подавления второй фазы OGD-индуцированного роста [Ca2+]i в нейронах и астроцитах в среднем на 35 % (p ≤ 0,05). Преинкубирование культуры с 3-ЕА (100 мкМ) приводит к дозозависимому снижению числа клеток на поздних стадиях апоптоза на 34 % (p ≤ 0,01). Возможным механизмом профилактики гибели нейронов и астроцитов в модели ишемии является повышение под влиянием 3-ЕА (100 мкМ) базовой и OGD/R-индуцированной экспрессии генов, кодирующих антиапоптотические белки BCL-2, STAT3, SOCS3 в среднем на 29 % (p ≤ 0,01), с параллельным подавлением транскрипции генов белков-регуляторов некроза и воспаления TRAIL, MLKL, Cas-1, Cas-3, IL-1β и TNFα в среднем на 26 % (p ≤ 0,01). Снижение под действием соединения 3-ЕА экспрессии генов, кодирующих моноаминоксидазы, и индукция транскрипции гена каталазы могут указывать на регуляцию базового и OGD/R-индуцированного окислительного статуса клеток коры головного мозга за счет биоэнергетики митохондрий. Наиболее выраженные эффекты соединение проявляет в концентрации 100 мкМ. Соединение 3-ЕА может рассматриваться как потенциальное церебропротекторное лекарственное средство.
