Изучение локализации мезенхимальных стволовых клеток костного мозга (МСК КМ), меченных зеленым (DiO C18) и красным (Rhod Chol) витальными флуорохромами, после их внутривенного (ВВ) и интрацеребрального (ИЦ) введения в организм крыс, у которых моделировали паркинсоноподобный синдром (ПС).Материалы и методы. Животные распределены на 3 группы по 6 особей в каждой: I -контрольная, ВВ и ИЦ введение меченых МСК КМ; II -модель ПС и ВВ введение зеленых МСК КМ; III -модель ПС и ИЦ введение красных МСК КМ. Трансплантацию клеток проводили на 7-е сутки моделирования ПС. Эффективность трансплантации МСК оценивали по степени уменьшения выраженности двигательных расстройств, локализацию меченных флуорохромами клеток в органах крыс выявляли на криосрезах головного мозга, легких, селезенки, сердца, почек и печени с помощью люминесцентного микроскопа через 4 и 9 сут после введения МСК.Результаты. Флуоресцентные прижизненные красители DiO C18 (зеленый) и Rhod Chol (красный) могут быть эффективно использованы для изучения свойств МСК в культуре и их миграции в организме подопытных животных. Установлена избирательная миграция трансплантированных меченых МСК КМ к зонам повреждения головного мозга при моделировании ПС у крыс.Выводы. Для доставки МСК в поврежденную зону головного мозга могут быть использованы их как ИЦ, так и менее инвазивное ВВ введение.Ключевые слова: паркинсоноподобный синдром, мезенхимальные стволовые клетки костного мозга, трансплантация, меченные флуорохромами клетки, клеточная миграция, эксперимент.
The effects of stress induced by hypothermia (+4 degrees C for 3 h) on the pathways of serotonin metabolism in the pineal gland and on its structure were studied in adult male Wistar rats. These experiments showed that the melatonin-forming function of the epiphysis undergoes phasic changes during adaptation: there was a significant increase during the first 15 min, which was followed by gradual inhibition (to initial by 30 min) and then sharp suppression (at 3 h). Suppression of the functional activity of the pineal gland occurred because of exclusion of a proportion of pinealocytes from the process of active functioning.
The effect of injection of exogenous melatonin on formation of the nocturnal peak of the melatonin concentration and intrapineal hormonogenesis was studied on mature male rats. It was shown that 3H-melatonin is selectively uptaken by pinealocytes, and the uptake intensity depends on the degree of saturation of an organism with exogenous melatonin. Exogenous melatonin exerts an inhibitory influence on the hormone self-production, at the same time apparently stimulating synthesis of epiphyseal peptides. The results can be considered an indication of the existence of an ultra-short connection between the mechanisms of production of epiphyseal indoles and peptides, which apparently plays an important role in autocrine regulation of the epiphyseal functions.
The article analyzes experimental data of histological examination of soft tissues of rabbits in the foreign body area after different types of wounds in dynamics. The effect of the temperature factor on the features of regeneration of connective tissue structures of the foreign body capsule is shown. Histological changes in the foreign body capsule in case of a vicious loop of maturation of connective tissue are detailed.
