Описана методика получения устойчивого конъюгата наночастиц оксида железа и цисплатина, определено оптимальное соотношение концентраций наночастиц и цисплатина. Проведен сравнительный анализ цитотоксического действия наночастиц, нагруженных цисплатином, и чистого цисплатина, без и при внешнем рентгеновском облучении клеток линии HeLa. Показано, что рентгеновское облучение клеток линии HeLa усиливает цитотоксическое действие конъюгата наночастицы с цисплатином на 35 % при p < 0,05, но не цисплатина в свободной форме.
Эффективность фотон-захватной терапии (ФЗТ) в значительной степени определяется свойствами используемого препарата, содержащего химический элемент, эффективно поглощающего внешнее рентгеновское излучение и выполняющего роль дозоповышающего агента (ДПА). Проводится сравнение эффективности висмута и гадолиния как ДПА в технологии ФЗТ с рентгеновским излучением 110 кВ для терапии поверхностных опухолей. Висмут и гадолиний сравниваются в одинаковой химической форме — в виде комплекса с диэтилентриаминопентауксусной кислотой, имеющего для обоих элементов похожие физико-химические характеристики. Исследование проведено на мышах с трансплантированной меланомой B16F10 в качестве опухолевой модели. Введение обоих ДПА осуществляли интратуморально в одинаковой дозе 5 мг ДПА на животное. Облучение проводили на рентгеновском аппарате с напряжением 110 кВ в дозе 20 Гр. Полученные результаты показывают существенно большую противоопухолевую эффективность ФЗТ как с гадолинием, так и с висмутом, по сравнению с близкофокусной рентгенотерапией. По критерию логарифма погибших клеток (lgN) наблюдалось увеличение значения lgN с 0,78 для близкофокусной рентгенотерапии до 2,5 для ФЗТ с гадолинием или висмутом при одинаковой величине экспозиционной дозы. Достоверных различий в противоопухолевой эффективности ФЗТ с висмутом и гадолинием для рентгеновского излучения 110 кВ не наблюдалось.
Бинарная лучевая терапия (БЛТ) — это разновидность лучевой терапии, использующая для наведения ионизирующего излучения на мишень специальные препараты. Как правило, используемый препарат не обладает выраженной собственной биологической активностью. В основе поражающего действия БЛТ лежит взаимодействие вторичного ионизирующего излучения с биологическими структурами. В составе специального препарата, способного избирательно накапливаться в опухоли, имеются химические элементы, которые поглощают внешнее ионизирующее излучение существенно более эффективно, чем химические элементы, составляющие живые ткани. В результате избирательного взаимодействия внешнего излучения с такими препаратами, находящимися в опухоли, происходит увеличение поглощенной дозы на величину от нескольких процентов до 5 раз. В настоящее время существует 2 разновидности БЛТ — это нейтрон-захватная терапия (НЗТ), использующая нейтронное излучение, и фотон-захватная терапия (ФЗТ), использующая рентгеновское излучение. НЗТ потенциально обладает большей терапевтической эффективностью, чем ФЗТ, однако стоимость необходимого оборудования и проведения процедуры ФЗТ существенно меньше и может быть реализована в условиях существующих клинических учреждений. Многочисленные исследования эффективности БЛТ, проведенные во многих странах мира, показывают потенциальную эффективность данной технологии в лечении злокачественных новообразований. Внедрение БЛТ в клиническую практику позволит существенно повысить противоопухолевую эффективность лучевой терапии и снизить количество фракций облучения вплоть до единственной. Потенциал БЛТ может быть полностью раскрыт лишь при разработке специализированных препаратов, удовлетворяющих требованиям БЛТ.
