Matrix metalloproteinases - enzyme complex necessary for maintenance of the homeostasis. He is a participant of normal, constantly current reconstruction of all living tissues. Action of pathogenic factors breaks harmonious work of this complex. Often violation is expressed by the excessive activity of enzymes amplifying pathogenic action. However and healing, which is accelerated new growth of tissue elements, can happen only at raised, compared with norm, metalloproteinase activity. Such situation demands from medicine of ability participate reasonably in work of enzyme system. The article presents some of the results of these actions.
Today, the brain-derived neurotrophic factor (BDNF) is a very popular substance. The high interest to this protein is due to its participation in key processes for life and health, including nervous regulation, carbohydrate and lipid metabolism, and exercise capacity. BDNF regulates preventive and therapeutic efficacy of exercise, emotional state, stress resistance, neuroplasticity, and neurogenesis. BDNF determines the action of the most universal and often the most effective therapeutic means, physical exercise. Therefore, BDNF appears to be the substance of primary importance for a burning issue of current medicine, prevention and treatment of hypertension. In addition to its hypotensive effect, BDNF has proved to be beneficial in hypertension-induced cognitive decline, schizophrenia, memory impairment, and emotional disorders.
Life experience and numerous experimental and clinical data evidence beneficial effects of mobility and physical activity on maintaining human bodily and mental health. Moreover, many mental, neurological, and neurodegenerative diseases and conditions, such as stroke, brain trauma, and drug addiction, for which there are no effective pharmacological therapies, can be prevented, considerably alleviated or slowed by physical exercise. Modern neurology has identified a number of mechanisms, by which muscular movements provide preventive and curative effects, including synthesis of neurotransmitters, neurotrophins and other growth factors, stimulation of neuroplasticity, formation of new and recombination of old connections, angiogenesis, mitohormesis, and neurogenesis.
The most common noninfectious diseases of the modern world, including metabolic syndrome, type 2 diabetes, chronic respiratory diseases, cerebrovascular and cardiovascular diseases, Alzheimer’s disease and other dementias, depression, tumors, sarcopenia, and osteoporosis, share a mechanism of homeostatic disorders, oxidative stress. Another common feature of these diseases is the old and senile age of patients. These characteristics are linked with impaired adaptability, which develops with older age. In principle, this combination is inevitable but the environment can influence it like all life. The article presents some of the most well-known techniques for creating such an environment (lifestyle) that could exclude or postpone (often for years) the onset of disease.
A research group at the Harvard University in 2013 and the next years has published a series of articles focusing on one of transforming growth factors b, GDF-11. According to these reports mammalian blood and tissue concentrations of GDF-11 decrease with age. Increasing the GDF-11 content in old mice using daily injections of recombinant GDF-11 or one-month parabiosis induced in young mice significantly reduced and even eliminated adverse, old age-specific changes in the heart, skeletal muscles, and brain. The reports of the Harvard group have attracted much attention of the scientific community, and were cited in hundreds of publications where GDF-11 was often called the protein of youth. The solidity of the Harvard team statement warranted solidity of test experiments, which used the most reliable methods. These experiments have expanded the knowledge of GDF-11 but did not confirm its right to be called a protein of youth.
scite is a Brooklyn-based organization that helps researchers better discover and understand research articles through Smart Citations–citations that display the context of the citation and describe whether the article provides supporting or contrasting evidence. scite is used by students and researchers from around the world and is funded in part by the National Science Foundation and the National Institute on Drug Abuse of the National Institutes of Health.
